Caracterização genética de mulungu (Erythrina velutina Willd.) em áreas de baixa ocorrência

Knowledge of the genetic diversity of the threatened tree species in the Brazilian semiarid region may contribute to the creation of effective strategies for their preservation and future use. The aim of this study was to select ISSR (Inter-Simple Sequence Repeat) primers capable of detecting genetic polymorphism among Mimosa caesalpiniaefolia Benth. individuals for a study of the genetic diversity of this species. A total of 27 ISSR primers were tested for their ability to amplify genomic DNA of three individuals (bulk); all of them amplified regions between the simple sequence repeats of the genome, however seven ISSR primers showed a good standard of amplification compared to the others. The seven primers selected were used to amplify the genomic DNA of nine individuals and generated a total of 78 loci, of which 52.7% were polymorphic. The content of polymorphic information of the primers ranged from 0.261 to 0.489, showing that selected primers are moderately informative; bootstrap analysis determined that 51 loci were enough to estimate the genetic diversity of the samples of individuals. We concluded that the markers generated by the primers UBC 807, UBC 824, UBC 827, UBC 840, UBC 851, UBC 873 and UBC 881 allow detection of the genetic polymorphism among individuals of M. caesalpiniaefolia, being useful to determine the genetic diversity of this species.Key words: semiarid, forest species, primers, polymorphism. ABSTRACT. Hyptis pectinata, popularly known as 'sambacaitá' or 'canudinho', is a medicinal and aromatic species widely used in the Brazilian Northeast. In Sergipe, the excessive extraction of natural resources may reduce the genetic variability of native plants. Thus, molecular markers have frequently been applied to the characterization of genetic diversity as the basis for germplasm conservation and breeding programs. The objective of the present study was to evaluate the genetic diversity of H. pectinata plants collected in different municipalities of the State of Sergipe using ISSR molecular markers. Thirty-four primers were tested, nine of which were selected for providing reproducible and analyzable amplification products, resulting in 67 polymorphic bands. The expected heterozygosity ranged from 0.32 to 0.45, with a mean of 0.39. Polymorphism information content was of 0.49, which classifies the markers as moderately informative. ABSTRACT. Erythrina velutina Willd. (Fabaceae: Papillionoideae) is a pioneer species found in tropical and subtropical regions of the world that has medicinal properties and that is used in reforestation projects. This species is rare in some areas of northeastern Brazil. This study aimed to characterize and compare genetic structures of natural and restored populations of E. velutina, with a focus on the selection of tree seeds. A total of 108 individuals from five natural populations and one restored population were analyzed using ISSR markers, resulting in 407 polymorphic fragments. A high rate of polymorphism was observed in the restored population. The highest genetic variability was identified within populations (82%). Genetic bottleneck tests were significant for the Carmópolis/Rosário do Catete and Laranjeiras natural populations along with the Laranjeiras restored population. Genetic distances significantly correlated with spatial distance. Only the restored population retained unique alleles. Similarly, increased genetic distance was observed in individuals of the restored populations compared to the other populations. Observed genetic variation in both natural and restored populations of E. velutina was moderate, thus enabling selection of divergent trees from those trees supplying seeds. Environmental protection and management of these areas is necessary for the maintenance of these individuals and subsequent reproduction. We recommend suggestions for E. velutina conservation, since the restoration model adopted in this study did not promote the development of the specimens until the reproductive stage in a fashion that aims to augment the soil seed bank supply, as is suggested for pioneer species.
