The space-time conservation element and solution element (CESE) method is applied to simulate the ITP and zone electrophoresis (ZE) separation phenomena. The CESE method expresses the governing equation in the integral form of the conservation law, and has a second-order accuracy in both space and time. The current results show that the CESE solutions for the ITP and ZE phenomena are more accurate than those obtained using conventional numerical schemes, which are characterized by serious numerical diffusion and oscillation. Furthermore, the CESE method suppresses the numerical oscillations or peaks observed in the results obtained using traditional second-order finite difference schemes. Finally, the results reveal that the CESE method accurately models the sharp boundaries between adjacent ITP samples under steady-state conditions. Overall, the results presented in this study demonstrate the numerical accuracy of the CESE method and confirm its applicability to the modeling of a range of electrophoretic phenomena.

An analysis of recent trends indicates that CE can show real advantages over chromatographic methods in ultratrace enantioselective determination of biologically active compounds in complex biological matrices. It is due to high separation efficiency and many applicable in-capillary electromigration effects in CE (countercurrent migration, stacking effects) enhancing significantly (enantio)separability and enabling effective sample preparation (preconcentration, purification, analyte derivatization). Other possible on-line combinations of CE, such as column coupled CE-CE techniques and implementation of nonelectrophoretic techniques (extraction, membrane filtration, flow injection) into CE, offer additional approaches for highly effective sample preparation and separation. CE matured to a highly flexible and compatible technique enabling its hyphenation with powerful detection systems allowing extremely sensitive detection (e.g. LIF) and/or structural characterization of analytes (e.g. MS). Within the last decade, more as well as less conventional analytical on-line approaches have been effectively utilized in this field and their practical potentialities are demonstrated on many new application examples in this article. Here, three basic areas of (enantioselective) drug bioanalysis are highlighted and supported by a brief theoretical description of each individual approach in a compact review structure (to create integrated view on the topic), including (i) progressive enantioseparation approaches and new enantioselective agents, (ii) in-capillary sample preparation (preconcentration, purification, derivatization), and (iii) detection possibilities related to enhanced sensitivity and structural characterization.

Progress achieved between 2014–2017 in the extraction and sample preparation of nucleic acid by isotachophoresis is reviewed in this paper. The isolation and purification of nucleic acids is very often compromised by a complex matrix such as blood and other bodily fluids, samples from the scene of crime, fossil samples, etc. While most of the common nucleic acids isolation techniques are based on extraction with inherent limitations with regard to quantitative results, isotachophoretic focusing is a quantitative process with a theoretically unlimited concentration factor. Since isotachophoresis belongs to less traditional approaches of nucleic acids purification, we present not only the latest developments in the application of isotachophoresis for the nucleic acids concentration but also a brief description of the principles of this method.
