Candidate Reference Method for the Quantification of Circulating 25-Hydroxyvitamin D3 by Liquid Chromatography–Tandem Mass Spectrometry

Abstract: Quantification of 25-hydroxyvitamin D 3 (25-hydroxycholecalciferol) in serum is the best-established indicator of vitamin D status (1 ). Vitamin D 3 (cholecalciferol) is absorbed from the diet, and, given sufficient ultraviolet irradiation, nutritionally adequate amounts of vitamin D 3 are formed in the skin from its precursor, 7-dehydrocholesterol. In the liver, vitamin D 3 undergoes hydroxylation to 25-hydroxyvitamin D 3 , which is further metabolized in the kidney to form the active metabolite 1,25-dihydro… Show more

“…[10] Another large study including 310 genotype 1 patients also found no association. [24] In 2 studies that frankly showed no association of response with vitamin D level [our current study and Kitson et al,2013] vitamin D levels were measured by LC-MS/MS methodology, [10,32] avoiding fallacies that may be induced by other commercially available kits. Another noticeable difference between the studies is the prevalence of advanced fibrosis.…”

“…The Elecsys assay is standardized against a modified version of the LC-MS/MS method published by Vogeser et al (22) with traceability to the reference material NIST SRM 2972, which consists of two ethanolic solutions of either 25-OHD2 or 25-OHD3 that are used to calibrate chromatographic methods (5,21,30). In addition, the performance of the in house LC-MS/MS was monitored by using NIST SRM 972 (vitamin D in human serum), which is the first certified reference material for quality-control use with officially assigned target values (5,18,19,30).…”

“…The Penzberg LC-MS/MS method utilizes an HPLC system (Agilent 1100 series) with additional binary pump for online-SPE (solidphase extraction) in conjunction with a triple quadrupole mass spectrometer with APCI source (atmospheric pressure chemical ionization; Thermo Scientific TSQ Quantum Ultra EMR). The calibration is performed against NIST standards as described above, using a modified version of a previously published LC-MS/MS method (22). The HPLC system was calibrated with the Chromsystems Serum Calibration Standard D3/D3 nr 38033.…”

“…None of the commercial immunoassays are, therefore, aligned to SRM 972. Several candidate reference methods for accurate and sensitive 25-OHD measurement have also been published in the recent years (19)(20)(21).…”
