Brain metabolomics study for the protective effects of Rhodiola crenulata extract on Alzheimer's disease by HPLC coupled with Fourier transform‐ion cyclotron resonance mass spectrometry

Abstract: In order to investigate the protective effects of Rhodiola crenulata extract on Alzheimer's disease, a brain metabolomics study in rats was conducted by high performance liquid chromatography coupled with Fourier transform ion cyclotron resonance mass spectrometry. Rat model was constructed by bilateral hippocampal injection of amyloid‐β peptide and immunohistochemistry was performed to evaluate the pharmacological effect of Rhodiola crenulata extract. Multivariate statistical analysis was used to discover pot… Show more

“…FT-ICR MS can determine the mass-to-charge ratio (m/z) of ions by detecting the frequency of their cyclotron motion in a high magnetic field and can use to analyze complex components, due to their high resolution and quality accuracy [27]. In recent years, it has been applied to decisively identify the chemical constituents by accurate mass and isotopic peaks in many TCMs, such as Cortex Fraxini, Rhodiola crenulata, Gansuibanxia decoction, and so on [28][29][30]. Therefore, HPLC-FT-ICR MS is a suitable method for the detection and characterization of chemical constituents in TCMs.…”

“…However, these single-target drugs are not only associated with various adverse effects and toxicity but also less effective in reversing the pathological progression of AD [ 29 , 30 ]. By contrast, because of their multitarget and multichannel properties, medicinal herbs and their derived ingredients (MHDIs) demonstrate considerably exceptional AD treatment outcomes compared with the aforementioned drugs [ 11 , 31 ]. Therefore, developing potential MHDI-based AD treatment strategies is highly warranted.…”

“…The patients show cognitive dysfunction, gradual memory decline, behavior abnormality, and social disorder [3], and cannot take care of themselves when the symptoms become severe, which brings great burden to the family and society. Therefore, it is crucial that find the means to alleviate the symptoms of the disease and to slow its progression [4].…”
