BmK CT enhances the sensitivity of temozolomide-induced apoptosis of malignant glioma U251 cells invitro through blocking the AKT signaling pathway

Abstract: Temozolomide (TMZ) is a drug that has been demonstrated to improve the survival time of patients with glioblastoma multiforme (GBM) when administered with concomitant radiotherapy. However, chemoresistance is one of the major obstacles in the treatment of GBM. In the present study, an MTT assay and flow cytometry were used to demonstrate that chlorotoxin-like toxin in the venom of the scorpion Buthus martensii Kirsch (BmK CT) markedly inhibited cell proliferation and induced apoptosis in U251 cells when combin… Show more

“…Due to the overexpression of phosphory-AKT, an increasing number of chemotherapy-resistant cases have been reported clinically [33]. Inhibitors of the PI3K-AKT signaling pathway, have identified to induce apoptosis of glioma cells and enhance the cytotoxicity of chemotherapy [33][34][35][36].…”

“…The present results demonstrated that VX-680 significantly decreased the expression of VEGFA. AKT is a type of serine/threonine protein kinase and serves an important role in cell proliferation and migration (27,28). The activation of the AKT signaling pathway can stimulate angiogenesis (18,20).…”

“…Glioma is a kind of CNS tumor developed from the deterioration of glial cells, which is one of the tumors with the highest degree of human evil (Molinaro et al, 2019). At present, the main treatment T A B L E 4 (Continued) (Continues) methods for glioma are surgical resection, chemotherapy, and radiotherapy; however, due to its invasive growth, it is difficult to be completely removed by surgery, and radiotherapy and chemotherapy usually achieve poor results and are often accompanied by serious side effects (Du et al, 2018;Suehiro et al, 2018). In recent years, the discovery of natural antitumor monomer drugs, such as paclitaxel and ginsenoside, made the anticancer effect of natural constituents to be gradually paid more and more attention (Kim et al, 1999;Yu et al, 2015).…”
