A microwave (MW)-assisted synthesis method for preparation of fluorescent BSA-stabilized and HSAprotected gold nanoclusters (BSA-AuNCs and HSA-AuNCs) has been developed. The reaction time can be shortened from tens of hours to several minutes, thanks to the superheating and non-thermal effects of the MW energy. The optimal experimental conditions including concentrations of BSA, HAuCl 4 and NaOH and the MW programme are investigated in detail. The as-synthesised BSA-AuNC product is well characterized by UV-vis spectroscopy, fluorescence spectroscopy, HRTEM, XPS, and IR spectroscopy. The HSA-AuNCs display strong red emission which is efficiently quenched by nitrogen oxides (NO x ), demonstrating its great potential in determining the intracellular concentration of NO x . The proposed MW-assisted synthesis method is simple, fast and should be applicable to prepare various protein-or enzyme-protected metal nanoclusters.
Background: Coronavirus disease 2019 (COVID-19) is a new respiratory and systemic disease caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection. The purpose of the present study was to investigate the association between cytokine profiles and lung injury in COVID-19 pneumonia. Methods: This retrospective study was conducted in COVID-19 patients. Demographic characteristics, symptoms, signs, underlying diseases, and laboratory data were collected. The patients were divided into COVID-19 with pneumonia and without pneumonia. CT severity score and PaO 2 /FiO 2 ratio were used to assess lung injury. Results: 106 patients with 12 COVID-19 without pneumonia and 94 COVID-19 with pneumonia were included. Compared with COVID-19 without pneumonia, COVID-19 with pneumonia had significantly higher serum interleukin (IL)-2R, IL-6, and tumor necrosis factor (TNF)-α. Correlation analysis showed that CT severity score and PaO 2 /FiO 2 were significantly correlated with age, presence of any coexisting disorder, lymphocyte count, procalcitonin, IL-2R, and IL-6. In multivariate analysis, log IL6 was the only independent explanatory variables for CT severity score (β = 0.397, p < 0.001) and PaO 2 /FiO 2 (β = − 0.434, p = 0.003). Conclusions: Elevation of circulating cytokines was significantly associated with presence of pneumonia in COVID-19 and the severity of lung injury in COVID-19 pneumonia. Circulating IL-6 independently predicted the severity of lung injury in COVID-19 pneumonia.
Breast cancer is the most common malignancy for women worldwide, while Triple Negative Breast Cancer (TNBC) accounts for 20% in all patients. Compared with estrogen receptor positive breast cancer, which could be effectively controlled via endocrine therapy, TNBC is more aggressive and worse in prognosis. It is therefore urgent and necessary to develop a novel therapeutic strategy for TNBC treatment. Recent studies identified Hippo signaling is highly activated in TNBC, which could be a driving pathway for TNBC progression. In our study, we determine RNF187 as a negative regulator for Hippo signaling activation. RNF187 depletion significantly decreases cell migration and invasion capacity in TNBC. These effects could be rescued by further YAP depletion. Depletion of RNF187 increases the YAP protein level and Hippo signaling target genes, such as CTGF and CYR61 in TNBC. Immuno-precipitation assay shows that RNF187 associates with YAP, promoting its degradation possibly via inducing YAP K48-dependent polyubiquitination. Interestingly, Our clinical data reveals that RNF187 reversely correlates with YAP protein level and Hippo target genes. RNF187 tends to correlate with good prognosis in TNBC patients. Our study provides evidence to establish a proteolytic mechanism in regulation Hippo signaling activation in TNBC.
We have previously demonstrated that upregulation of Sonic hedgehog (SHH) expression in allergic airway epithelia essentially contributes to the goblet cell metaplasia and mucous hypersecretion. However, the mechanism underlying the upregulation of SHH expression remains completely unknown. In cultured human airway epithelial cells, IL-4/IL-13 but not IL-5 robustly induces the mRNA and protein expression of SHH and in turn activates SHH signaling by promoting the JAK/STAT6-controlling transcription of SHH gene. Moreover, intratracheal instillation of IL-4 and/or IL-13 robustly activates STAT6 and concomitantly upregulates SHH expression in mouse airway epithelia, whereas, in Club cell 10-kDa protein (CC10)-positive airway epithelial cells of children with asthma, activated STAT6 closely correlates with the increased expression of SHH and high activity of SHH signaling. Finally, intratracheal inhibition of STAT6 by AS-1517499 significantly diminished the allergen-induced upregulation of SHH expression, goblet cell phenotypes, and airway hyperresponsiveness, in an ovalbumin- or house dust mite-induced mouse model with allergic airway inflammation,. Together, upregulation of SHH expression by IL-4/IL-13-induced JAK/STAT6 signaling contributes to allergic airway epithelial remodeling, and this study thus provides insight into how morphogen signaling is coordinated with Th2 cytokine pathways to regulate tissue remodeling in chronic airway diseases.
In this study, the cationic Poly[Styrene-Butyl acrylate-(P-vinylbenzyl trimethyl ammonium chloride)] P(St-BA-VBT) nanospheres with N+(CH3)3 functional groups were successfully synthesized by soap-free emulsion polymerization and applied to different fabrics by pad-cure process. After the pad-cure process, the nanospheres were deposited on the surface of the modified cotton fibers successfully without forming a continuous film structure. The X-ray Photoelectron Spectroscopy (XPS) and the Fourier transform infrared (FTIR) results demonstrated that P(St-BA-VBT) nanospheres were adsorbed on the surface of cotton fibers successfully. The excellent color strength value and outline sharpness of the plain, twill, and honeycomb fabrics can be achieved when the nanosphere concentration, sodium bicarbonate, and steaming time were 1 g/L, 10 g/L, and 6 min, respectively. The plain fabrics exhibited the smallest color strength (K/S) values and the best outline sharpness, followed by twill and honeycomb fabrics, which displayed the largest K/S values and the worst outline sharpness after the same treatment. Besides, all the three fabrics showed excellent rubbing fastness and washing fastness. The cationic P(St-BA-VBT) nanospheres modification of the cotton fabrics provides a novel potential approach to obtain good printing efficiency without affecting the tensile breaking strength of cotton fabrics significantly.
