Biotin-Streptavidin Enzyme-linked Immunosorbent Assay for the Determination of Dibutyl Phthalate in Beverages and Drinking Water Using a Specific Polyclonal Antibody

Abstract: A specific polyclonal antibody targeting dibutyl phthalate (DBP) was obtained, and a sensitive indirect competitive biotin-streptavidin enzyme-linked immunosorbent assay (BA-ELISA) was developed for the determination of DBP in beverages and drinking water. Under optimal conditions, good linearity was achieved within a range of 0.02 to 8.97 μg·L −1 . The limit of detection (IC 10 ) was 5 ng·L −1 , and the median inhibitory concentration (IC 50 ) was 0.36 μg·L −1 . The BA-ELISAwas highly selective, with lower cr… Show more

“…Compared with the sensitive biotin-streptavidin-based enzyme-linked immunosorbent assay (BA-ELISA, Sun & Zhuang, 2015), which has also been studied by our research team, the enzyme chromogenic reaction signal used in the BA-ELISA was replaced by fluorescent signal labelled on template DNA in rt-IPCR analysis. Multiple-fold magnification of fluorescent signal can be achieved with the amplification of template DNA.…”

“…On the basis of above, the biotin-streptavidin system has introduced an enzyme-based immunoassay to amplify reaction signal because the biotin-streptavidin enzyme-linked immunoassays (BA-ELISA) has demonstrated that eight-folds higher sensitivity than the traditional ELISA under the same antibody and coating antigen (Wang, Zhang, Gao, Duan, & Wang, 2010). This strategy has been reported by the Zhuang group, including the biotin-streptavidin enzyme-based immunoassay for the determination of PAEs mainly in beverages and drinking water, milk and milk products and wine samples (Sun & Zhuang, 2014, 2015a, 2015b, 2015c.…”

“…Phthalic acid esters, as a class of plasticizers, are often added to plastic food-packaging materials to increase the plasticity and ductility of the plastic (Benjamin, Pradeep, Josh, Kumar, & Masai, 2015;Sørensen, 2006;Sun, & Zhuang, 2015a). However, such plasticizers tend to migrate into foods as a result of environmental factors such as storage temperature and food polarity (Keresztes, Tatar, Záray, & Czegeny, 2013;Salazar-Beltrán, Hinojosa-Reyes, Ruiz-Ruiz, Hernández-Ramírez, & Guzmán-Mar, 2018;Shi et al, 2012).…”
