Biodistribution and toxicity of MR imaging contrast media

Abstract: ISIGNAL INTENSITY on a magnetic resonance (MR) image is mainly determined by proton density and T1 and T2. Image contrast, described as the difference in signal intensity between two tissues, is dependent on a number of physical parameters and can be altered by changing these parameters. Contrast agents are pharmaceuticals that can accentuate the difference in signal intensity between two adjacent tissue compartments by directly interfering with the proton relaxation times in one more than in the other. Such a… Show more

“…This may suggest either that saturation does not occur in humans or that it occurs at a similar number of multiple GBCA administrations. Animal studies are not strictly Data represent analysis of signal intensity ratios using the GAM function DN/MCP dentate nuclei-to-middle cerebellar peduncle ratio, Previous previous administration of gadodiamide, MRIs number of MRIs performed, GAM generalized additive models equivalent to human studies; for example, the excretion halflife of GBCAs in healthy rats [20] is markedly shorter than in humans [21], thereby reducing tissue exposure [10] which may suggest that saturation effects may be observed in humans with lesser numbers of GBCA administrations.…”

“…injection, low molecular weight Gd chelates are rapidly eliminated through glomerular filtration, with a blood half-life shorter than 30 min. 25,26 Similarly, lanthanide-based CAs, which are released in the interstitium in the case of labeled cell death, also ought to be rapidly cleared by the kidney. With this class of CA, label recapture by other cells can be expected to be less of a problem than with SPIOs.…”

“…It is already known that in rats, renal excretion is much more rapid compared with humans (27). With a prolonged intravascular residence time, the delineation of ischemic myocardium might be even better.…”
