Benzofurazan‐based fluorophore for the selective determination of flupentixol dihydrochloride: Application to content uniformity testing

Abstract: One of the most commonly used drugs in treatment of schizophrenia is flupentixol dihydrochloride, therefore it is important to develop a simple, low cost and sensitive spectrofluorimetric method for the estimation of flupentixol dihydrochloride. The yellow fluorescent product that is generated from the nucleophilic substitution reaction of the free lone pair of the alcoholic hydroxyl group of the drug and 4-chloro-7-nitrobenzofurazan (NBD-Cl) in Mcllvaine buffer pH 7.0 was estimated at 510 nm (λ 460 nm). The v… Show more

“…NBD-Cl was previously used as a fluorogenic reagent for determination of many pharmaceutical drugs having primary or secondary amine moiety at mild alkaline medium producing fluorogenic products. [15][16][17][18] In this work, the reaction of secondary amine moiety in TIGE with NBD-Cl was achieved using borate buffer at pH 9 ± 0.2 at 75°C after heating for 15 min. The proposed method showed higher sensitivity than the previously reported spectrofluorimetric methods.…”

“…Highly sensitive, selective, and accurate bioanalytical methods are required to undertake pharmacological and toxicological investigations and clinical TDM of antipsychotics, as well as to address the problems associated with polypharmacy and drug metabolism (9). When the literature studies are examined, it has been seen that there are voltammetric (10), potentiometric (11), spectrofluorimetric (12), second-order spectrophotometric (13), reversed-phase high performance liquid chromatographic (HPLC) (14), LC-MS/MS (3) and LC-ESI-MS (15) methods designed to determine FLP from the pharmaceutical preparations and biological samples in an individual manner.…”
