The continuous use of synthetic anthelmintics against gastrointestinal nematodes (GINs) has resulted in the increased resistance, which is why alternative methods are being sought, such as the use of natural products. Plant essential oils (EOs) have been considered as potential products for the control of GINs. However, the chemical composition and, consequently, the biological activity of EOs vary in different plant cultivars. The aim of this study was to evaluate the anthelmintic activity of EOs from cultivars of Ocimum basilicum L. and that of their major constituents against Haemonchus contortus. The EOs from 16 cultivars as well the pure compound linalool, methyl chavicol, citral and eugenol were used in the assessment of the inhibition of H. contortus egg hatch. In addition, the composition of three cultivars was simulated using a combination of the two major compounds from each. The EOs from different cultivars showed mean Inhibition Concentration (IC50) varying from 0.56 to 2.22 mg/mL. The cultivar with the highest egg-hatch inhibition, Napoletano, is constituted mainly of linalool and methyl chavicol. Among the individual compounds tested, citral was the most effective (IC50 0.30 mg/mL). The best combination of compounds was obtained with 11% eugenol plus 64% linalool (IC50 0.44 mg/mL), simulating the Italian Large Leaf (Richters) cultivar. We conclude that different cultivars of O. basilicum show different anthelmintic potential, with cultivars containing linalool and methyl chavicol being the most promising; and that citral or methyl chavicol isolated should also be considered for the development of new anthelmintic formulations.

Gastrointestinal nematodes are responsible for great economic losses in sheep raising, and their control has long been carried out almost exclusively by the administration of anthelmintics, which have led to serious resistance problems. In the search for alternative control measures, phytotherapic research is highlighted. The aim of this study was to evaluate the action of Anethum graveolens (dill) essential oil on different stages of Haemonchus contortus life cycle, as well its cytotoxicity MDBK (Madin-Darby bovine kidney) cells. H. contortus larvae and eggs were obtained from infected sheep feces, and essential oil extracted from plant seeds through the Clevenger apparatus. 9.4, 4.7, 2.35, 1.17. 0.58 and 0.29 mg/mL concentrations were evaluated. The Egg Hatch Inhibition (HI), Larval Development Inhibition (LDI) and Larval Migration Inhibition (LMI) techniques were used. Thybendazole 0.025 mg/mL in HI and Levamisole 0.02 mg/mL in the LDI and LMI tests were used as positive controls, while distilled water and a Tween 80 solution were used as positive negative controls. The inhibition results obtained for the highest oil concentration were: HI 100%, LDI 98.58% and LMI 63.7%, differing ( <0.05) from negative controls. Main A. graveolens oil components present in 95.93% of the total oil were Dihydrocarvone (39.1%), Carvone (22.24%), D-Limonene (16.84%), Apiol (10.49%) and Trans-dihydrocarvone (7.26%). Minimum A. graveolens essential oil concentrations required to inhibit 50% (IC50) of egg hatching, larval development and larval migration were 0.006 mg/mL, 2.536 mg/mL and 3.963 mg/mL, respectively. Cell viability in MDBK (Madin-Darby bovine kidney) cells, when incubated with A. graveolens essential oil, was 86% for the highest (9.4 mg/mL) and 99% for the lowest concentration (0.29 mg/mL). A. graveolens essential oil, according to the results obtained in this study, is a promising alternative in sheep gastrointestinal nematode control. ResumoOs nematoides gastrintestinais são responsáveis por grandes perdas econômicas na ovinocultura, e seu controle tem sido realizado quase exclusivamente pela administração de anti-helmínticos, que levaram a sérios problemas de resistência. Na busca de medidas alternativas de controle, destaca-se a pesquisa fitoterápica. O objetivo deste trabalho foi avaliar a ação do óleo essencial de Anethum graveolens (endro) em diferentes estágios de Haemonchus contortus, bem como testar a viabilidade celular para o óleo. Larvas e ovos de H. contortus foram obtidos de fezes de ovinos infectados e óleo essencial extraído de sementes de plantas através do aparelho de Clevenger. As concentrações avaliadas foram 9,4, 4,7, 2,35, 1,17, 0,58 e 0,29 mg/mL. The present study aimed to evaluate the in vitro potential of aromatic plants Aloysia triphylla (cidró ou erva-luísa), Corymbia citriodora (eucalipto-limão), Lippia alba (falsa-melissa) e Piper gaudichaudianum (pariparoba), in helminths eggs and larvae, determine their main constituents. For this, the essential oils (OEs) of the plants were extracted from leafs, in the clevenger apparatus and to determine the main constituents was carried out the gas chromatography coupled to mass spectrometry. The Egg eclodibility assay (EEA) and Larval development test (DLT) were performed using essential oils at concentrations ranged from 75 at 0.2%. mg.mL, and all tests followed by commercial anthelmintic positive control and negative control distilled water. For EEA, A. triphylla e L. alba OEs demonstred equivalente efficacy to the commercial drug at all concentrations that was tested, while P. gaudichaudianum OE demonstred equivalente efficacy at 15 mg/mL and the C. citriodora OE at 10 mg/mL. About DLT, all concentratios of the studied essential oils were effective as the comercial drug results in this test. The dominant constituent in OEs were: geraniale (A. triphylla), citronellal (L. alba), linalool (L. alba) and α-humuleno (P. gaudichaudianum). Based on the results presented here, the OEs present potential for use as natural anthelmintics for the treatment of endoparasitosis in sheep.
