Gastrointestinal Nematode Infection (GIN) are the main constraint to the production of small ruminants. Studies of medicinal plants have been an important alternative in the effort to control these parasites. Therefore, the purpose of this study was to evaluate the in vitro ovicidal and larvicidal activity of essential oil of Rosmarinus officinalis. The oil was extracted, analyzed by gas chromatography and tested on GIN eggs and larvae in six concentrations, 227.5mg/mL, 113.7mg/mL, 56.8mg/mL, 28.4mg/mL, 14.2mg/mL and 7.1mg/mL. To determine the ovicidal activity, GIN eggs were recovered from sheep feces and incubated for 48h with different concentrations of the oil. For the evaluation of larval migration, third-stage larvae (L3) were obtained by fecal culture, and associated with the essential oil for 24h at the same concentrations, after which they were left for another 24 hours on microsieves, followed by the count of migrating and non-migrating larvae. The assays of R. officinalis oil showed a significant (p<0.05) 97.4% to 100% inhibition of egg hatching and a significant (p<0.05) 20% to 74% inhibition of larval migration. The main constituent revealed by gas chromatography was Eucalyptol. The results indicate that R. officinalis essential oil has ovicidal and larvicidal activity on sheep GINs.
vulnerability. The fact that these people share the environment with animals promotes the establishment of zoonotic parasitic infections, as well as the resultant parasitic cycles. Thus, parasites present in the environment must be identified, so that control measures can be recommended. In this context, this study’s objective was to evaluate environmental contamination by parasitic forms in a socially vulnerable community in southern Rio Grande do Sul. A total of 100 soil samples collected from the community were processed by a sodium dichromate centrifuge-flotation technique and analyzed by a compound microscope (40X objective) for the identification of parasite eggs, oocysts and cysts. All points were positive for two or more parasites, with the identification of 33.59% non-identified coccidian oocysts, Strongylida (25.4%), Ascaridida (21.31%), Trichuris spp. (8.19%), Toxocara spp. (3.27%), Amoebas (4.08%), Dioctophyma renale (2.45%), and Giardia spp. (1.63%). The presence of parasitic forms in all points analyzed surpasses other studies of environmental contamination carried out in the southern region of Brazil. In addition, the identification of several parasitic forms with zoonotic potential is concerning, since it shows the possibility of parasitic transmission to humans and other animals. In view of the results, the conclusion is that the environment analyzed is contaminated by parasitic forms, constituting a serious public health problem. Therefore, implementing educational and preventive measures in the community to control parasites is of crucial importance.
RESUMOcontrole dos helmintos gastrintestinais em ovinos, realizado exclusivamente através de anti-helmínticos, tem como consequência a seleção de populações de helmintos resistentes a praticamente todos os fármacos disponíveis. Isso tem motivado à busca de alternativas para controle, entre as quais destacamos a utilização de fitoterápicos. Neste contexto, o objetivo deste estudo foi avaliar a atividade ovicida in vitro do óleo essencial e do extrato hidroalcoólico de Ocimum basilicum em nematódeos de ovinos e determinar os constituintes químicos majoritários do óleo essencial. Para os estudos in vitro foram utilizadas fezes de ovinos naturalmente infectados, testadas frente ao óleo essencial de O. basilicum nas concentrações de 32 a 0,25 mg/mL e ao extrato hidroalcoólico nas concentrações 40 a 1,25 mg/mL. Como controle positivo foi utilizado o princípio ativo tiabendazol (0,025 mg/mL) e negativo a água destilada. A análise química do óleo essencial determinou como constituintes majoritários chavicol (34,68%) e linalool (27,68%). O óleo essencial apresentou atividade ovicida acima de 99% em todas as concentrações testadas. O extrato hidroalcoólico nas concentrações entre 40 e 20 mg/mL obteve 100% de ação, decrescendo a partir de 10 mg/mL, atingindo 9,76% de atividade na concentração 1,25 mg/mL. Os controles, positivo e negativo, obtiveram 98,57% e 8,5%, respectivamente. O valor da concentração letal média (CL 50 ) do extrato hidroalcoólico e óleo essencial foram de 4,797 mg/mL e 0,2123 mg/mL, respectivamente. O. basilicum apresentou ação antihelmíntica in vitro sobre ovos de nematódeos gastrintestinais de ovinos, com maior potencial para o óleo essencial, constituindo um promissor fitoterápico para estudos visando à busca de moléculas bioativas para serem utilizadas no controle dos nematódeos gastrintestinais. Resumo Este estudo avaliou a ação in vitro e in vivo de extratos de Eugenia uniflora em nematódeos gastrintestinais de ovinos. No teste de inibição da eclodibilidade dos ovos, extratos aquosos e hidroalcoólicos foram testados nas concentrações de 40 a 1,25 mg/mL. O efeito citotóxico foi mensurado através do ensaio de MTT em células VERO nas concentrações de 4000 a 1,95 µg/mL. A composição química foi analisada através da marcha fitoquímica qualitativa. No teste in vivo, foram administrados 100 mg/kg, por via oral, do extrato hidroalcoólico por três dias em ovinos naturalmente infectados e realizada coleta de fezes para estimar a redução de ovos por grama de fezes (OPG). Diferentes extratos de E. uniflora inibiram a eclodibilidade dos ovos, com percentual de inibição variando de 14,56 a 99,75%, sendo os hidroalcoólicos mais promissores que os aquosos. Na marcha fitoquímica, compostos com ação anti-helmíntica comprovada foram observados, como flavonoides, saponinas, taninos e triterpenos. Nas condições testadas, os extratos apresentaram citotoxicidade nula a moderada. In vivo, a redução do OPG foi observada apenas no grupo controle (anti-helmíntico). Os extratos de E. uniflora mostraram-se promissores com ação in vitro, necessitando estudos que avaliem outras concentrações e formas de administração in vivo.
