Association of P2RX7 functional variants with localized aggressive periodontitis

Abstract: Objective The purpose of this study was to investigate involvement of the P2X7 receptor in the rare condition, localized aggressive periodontitis. Material and methods Peripheral blood from 220 African Americans (103 with localized aggressive periodontitis and 117 healthy unrelated controls) was stimulated with lipopolysaccharide from E coli and Porphyromonas gingivalis. P2RX7 single nucleotide polymorphisms rs208294 (H155Y), rs1718119 (T348A), rs2230911 (T357S) and rs3751143 (E496A) were genotyped in 103 loca… Show more

“…Thus, the decrease in GM-CSF levels seems beneficial in inflamed mucosa surfaces, including in periodontitis. Increased GM-CSF levels were associated with an allele of increased susceptibility to aggressive periodontitis ( Harris et al, 2020 ), whereas the destruction of the alveolar bone was dependent on GM-CSF in a mouse experimental model ( Lam et al, 2015 ).…”

“…One SNP in LD (r 2 = 1.0, D’ = 1.0) is a gain-of-function variant, rs1718119 ( https://ldlink.nci.nih.gov/ ). rs1718119 is associated with higher pain intensity scores in females with diabetic peripheral neuropathy ( Ursu et al., 2014 ) and with inflammatory conditions including systemic lupus erythematosus, chronic obstructive pulmonary disease, and localised aggressive periodontitis ( Chen et al., 2013 ; Dai et al., 2018 ; Harris et al., 2020 ). Homozygous carriage of the minor allele of rs1718119 is associated with an ATP-induced increase of IL-1β from monocytes and with increased levels of IL-6 in whole blood ( Harris et al., 2020 ; Stokes et al., 2010 ).…”

“…However, patients often ignore the disease in its early stages. Therefore, most of them have already experienced symptoms including looseness, displacement or loss of teeth by the time they visit the clinic, which has had a severe impact on their daily behaviors including chewing and vocalization, and posing difficulties and challenges for clinical diagnosis and treatment (11,12). Localized aggressive periodontitis has a high incidence in clinical practice, usually caused by a mixed infection of many kinds of anaerobic bacteria (13).…”
