Association between lysyl oxidase polymorphisms and oral submucous fibrosis in older male areca chewers

Abstract: The elder OSF patients were increased in LOX Arg158Gln. Our findings may suggest a potential application in risk population selection using LOX polymorphism for preventive intervention of OSF genesis in a subset of areca chewers.

“…66 Copper dependant enzyme lysyl oxidase is critical for collagen cross linking and organization of ECM. 67 Salivary copper is found to be higher in arecanut chewers. This finding indicates that soluble copper found in arecanut is released into the oral environment and its buccal absorption may contribute to fibrosis of oral tissues where copper is deposited.…”

“…The incidence rate of oral squamous cell carcinoma in patients with OSF, after a follow-up period of 17 years, has been estimated to be 7.6% (2). Areca-associated oral squamous cell carcinoma is the third most common malignancy in the developing world (3). Despite the fact that OSF is, at times, preceded by and/or linked to vesicle formation, it is known to be associated with a juxta-epithelial inflammatory reaction, which is followed by a fibroelastic change of the lamina propria, with epithelial atrophy resulting in stiffness of the oral mucosa and causing inability to eat and trismus (4).…”

“…As a result, LOX Arg158Gln could affect PCP cleaving efficiency and reduce the availability of the LOX active form. Recently, an association has been described between the G473A polymorphism and oral submucous fibrosis (a lesion characterized by fibrosis of the oral submucosa and the progressive tissue rigidity) in South Asian older males chewing areca nuts (62). Therefore, the possibility that LOX-dependent cardiac fibrosis is genetically determined requires further investigation.…”
