Assessments of Biodiversity Based on Molecular Markers and Morphological Traits among West-Bank, Palestine Fig Genotypes (Ficus carica L.)

Abstract: Both morphological characters and PCR-based RAPD approaches were used to determine the genetic diversity and relatedness among nine fig genotypes grown at the northern region of the West-Bank, Palestine. Although we tested 28 primers for the RAPD technique, only 9 produced reasonable amplification products. A total of 57 DNA loci were detected in which 70.2% were polymorphic. DNA fragments presented a minimum of 3 and a maximum of 9 polymorphic bands using primers OPT-10 and OPA-18, respectively. Primers exhib… Show more

“…Our results reflected similar findings as reported earlier in studies of the genetic variation in other species at the cultivar level by using RAPD markers (Singh et al, 2012;Kalpanaa et al, 2012;Badfar-Chaleshtori et al, 2012;Basheer-Salimia et al, 2012;Rajesha et al, 2013) and among Palestinian faba bean landraces (Basheer-Salimia et al, 2013).…”

“…The use of Random amplified polymorphic DNA (RAPD) technique offers a simple, fast, efficient and inexpensive method (Basheer-Salimia et al, 2012). Furthermore, it does not need knowledge of marker sequences and can produce abundant polymorphic DNA fragments (Kocsis et al, 2005 andAchtak et al, 2009).…”

“…The primers BC-261, -322, and -298 were the most useful RAPD primers to assess the genetic diversity since they revealed relatively high Rp rates (5.236, 3.618, and 3.150, respectively). In fact, higher Rp rates also establish the effectiveness of RAPD molecular markers in detecting polymorphism among plant species (Basheer-Salimia et al, 2012).…”

“…Compared with other molecular techniques, RAPD is based on random amplification of bases from short primers. The use of the RAPD technique offers a simple, fast, efficient, and inexpensive method (Basheer-Salimia et al, 2012). Furthermore, it does not require prior knowledge of the sequences of the markers and can produce abundant polymorphic fragments (Kocsis et al, 2005;Achtak et al, 2009).…”
