The genetic diversity of 13 local Palestinian fig genotypes was investigated using RAPD markers. Among the 30 tested primers, 28 revealed various banding patterns and 2 generated no polymorphic bands. In addition, 13 primers (46.4%) produced good amplification products with high intensity and pattern stability. A total of 94 DNA fragments (loci), separated by electrophoresis on agarose gel were detected, ranging in size from 190 to 1300 bp. Of these fragments, 72 (76.6%) were polymorphic and 22 (23.4%) were monomorphic. A minimum of three and a maximum of eight DNA fragments were obtained using (OPH-02 and OPT-10) as well as (OPA-13, OPA-18 and OPY-07) primers respectively. The maximum percentage of polymorphic markers was 100.0 (Z-5, Z-12, and OPT-10) and the minimum was 60.0 (OPH-02). Primers OPY-07 and OPA-13 revealed high collective resolving power (Rp) values with 4.640 and 4.760 respectively and therefore, they were the most useful RAPD primers to assess the genetic diversity in the Palestinian figs. Genetic distance matrix showed an average distance range from 0.186 to 0.559 with a mean of 0.373. Thus, the cultivars tested in this study were characterized by large divergence at the DNA level. To our knowledge, this is the first report using RAPD marker to assess genetic diversity of Palestinian figs.

Both morphological characters and PCR-based RAPD approaches were used to determine the genetic diversity and relatedness among nine fig genotypes grown at the northern region of the West-Bank, Palestine. Although we tested 28 primers for the RAPD technique, only 9 produced reasonable amplification products. A total of 57 DNA loci were detected in which 70.2% were polymorphic. DNA fragments presented a minimum of 3 and a maximum of 9 polymorphic bands using primers OPT-10 and OPA-18, respectively. Primers exhibited collective resolving power values (Rp) of 18.826. The Mwazi genotype showed the highest genetic distances among all of the other genotypes. Morphologically, considerable variations were found using 41 quantitative and qualitative traits. Adloni could be a very promising genotype for fresh consumption due to its very late maturation period, extended harvesting period, variable fruit size, and easy skin peeling. In addition, 7 genotypes presented firm fruits, which are a very important criterion for exporting purposes. Dendrogram constructed by UPGMA based on RAPD banding patterns appear somewhat contradictory to the morphological descriptors particularly with Swadi and Biadi genotypes (closed genetically and distanced morphologically), which might be attributed to the phenotypic modifications caused by environmental differences across regions. These preliminary results will make a fundamental contribution to further genetic improvement of fig crops for the region.

ABSTRACT. Until now, neither phenotypic nor molecular approaches have been used to characterize the landraces of Palestine faba beans (Vicia faba). We used PCR-based RAPD markers to determine the genetic diversity and relatedness among 26 Palestinian faba bean landraces (traditional farmers' varieties) from 8 localities in the West Bank, Palestine. In tests with 37 primers, 14 generated no polymorphic bands, 12 exhibited weak and unclear products, and 11 primers produced good amplification products with high intensity and pattern stability. Ninety-four DNA fragments (loci) were detected, with an average of 8.54 loci per primer and size ranging from 160 to 1370 bp. A minimum of 4 and a maximum of 14 DNA fragments were obtained using (OPA-05 and OPA-09) and (BC-261) primers, respectively. The maximum percentage of polymorphic markers was 71.4 (BC-298) and the minimum was 50.0 (OPA-05, -09, -16). The 11 primers exhibited Genetic diversity of faba bean genotypes using RAPD markers relatively high collective resolving power (Rp) values of 26.316, and varied from 0.154 for the OPA-09 primer to 5.236 for the BC-261, with an overall mean of 2.392. The primers BC-261, -322, and -298 were found to be the most useful RAPD primers to assess the genetic diversity of Palestinian faba beans, as they revealed relatively high Rp rates (5.236, 3.618, and 3.150, respectively). Based on the Jaccard coefficient, the genetic distance ranged from 0.358 to 0.069, with a mean of 0.213. We conclude that the RAPD technique is useful for determining genetic diversity and for developing suitable fingerprints for faba bean landraces grown in Palestine.

There is limited knowledge about the relationships of epiphytic microbiomes associated with the phyllosphere of different Vitis vinifera cultivars in the same vineyard and terroir. To address this research gap, we investigated the microbiome compositionof 36 grapevine genotypes grown in the same vineyard in different plant sections during the growing season. Using high-throughput NGS-based metagenomic analysis targeting the ITS2 and the V4 regions of the 16S ribosomal gene of fungal and bacterial communities, respectively, weassessed the impact of grapevine genotypes on microbial assemblages in various parts of the phyllosphere. The results indicated that different phyllosphere tissues display high microbial diversity regardless of the cultivars’ identity and use. The selected three phyllosphere parts representing three distinct phenological stages, namely bark and bud, berry set, and fruit harvest, had almost a similar number of fungal OTUs, while a difference was recorded for the bacterial species. The fruit harvest stage hosted the highest number of bacterial OTUs, whereas the bark and bud stage contained the lower. Bacterial dominant phyla were Proteobacteria, Bacteroidetes, Actinobacteria, and Firmicutes, and the genera were Gluconacetobacter, Erwinia, Gluconobacter, Zymobacter, Buchnera, Pseudomonas, Pantoea, Hymenobacter, Pedobacter, Frigoribacterium, Sphingomonas, and Massilia. For fungi,the dominant phyla were Ascomycota and Basidiomycota, and the genera were Aureobasidium, Cladosporium, Alternaria, Aspergillus, Davidiella, Phoma, Epicoccum, Rhodosporidium, Glomerella, Botryosphaeria, Metschnikowia, Issatchenkia, and Lewia. Both the genotype of the cultivar and the phenological stage appeared to considerably impact the shape of microbial diversity and structure within the same terroir. Taken together, these results indicate that microbiome analysis could be proved to be an important molecular fingerprint of cultivars and provide an efficient management tool for the traceability of wine and grape end products. Moreover, the unique identity of cultivars’ microbial signatures highlights the need for further development of precision management to support viticulture sustainability in the face of climate change.

Viticulture is globally an important economic activity, and grapevine microbiomes hold a significant role in influencing yield and quality. Earlier studies showed that cultivar and agronomic management affect grapevine microbiome structure and, potentially, the quality of the end product. While microbial dynamics and ecology were established on some grapevine tissues, i.e., leaves and grapes, there is less knowledge deciphering microbiomes on other tissues, i.e., barks and buds. Moreover, although the impact on the microbiome of the so-called “vitivinicultural terroir” is well established, there are limited data considering microbiomes of genetically diverse cultivars within the same environment. Our study aims to explore microbiome diversity on bud and bark tissues of 37 different grapevine cultivars under the same environment and agronomic management. We targeted the V2-9 regions of the 16S rRNA gene of the microbiomes in bark and buds at the onset of new vegetation and bud expansion using Ion Torrent sequencing technology. Our results show that these tissues display high bacterial diversity regardless of cultivars’ use. Proteobacteria, Bacteroidetes, and Actinobacteria were the most prevalent among 11 detected phyla. The genotype of the cultivar seems to affect bacterial diversity and structure (p < 0.001) within the same environment. Our approach highlights the efficiency of high-throughput sequencing to unfold microbiomes of several grapevine parts that could be an important source of microbial inoculation and an important molecular fingerprint of the wine and grape end products.
