Thermophilic microorganisms are quite attractive for the study of biodiversity and evolutionary process as well as biotechnological applications. These organisms provide significant advantages for industrial and biotechnological processes occurring fast and efficiently at high temperatures. Possible potential also is getting increase thanks to isolation of new strains, determination of new metabolites and their pathway. Within the scope of this study, thermophilic bacterial community was investigated with a combination of classical microbiology and molecular biology approaches including fluorescent in situ hybridization, amplified ribosomal DNA restriction analysis, and polymerase chain reaction of 16S rRNA gene. Archaea and Bacteria domain were screened by Fluorescence in situ Hybridization technique. At the end of culture-dependent methodology, Paenibacillus lactis E3.1 (MK573857), Brevibacillus borstelensis E3.2 (MK573871), Paenibacillus naphthalenovorans E2.2 (MK573627), Paenibacillus sp. E3.5 (MK573870) were obtained. Furthermore, these isolates were screened with regards to protease production capabilities. At the end of screening studies, the highest protease activity (300 U/mg) was observed for Paenibacillus lactis E3.1.

Microorganisms inhabit extreme environments are novel in terms of phylogeny and also valuable source of extracellular hydrolytic enzymes. The present study focused on isolation and characterization of moderately thermophilic heterotrophic bacteria from Thar Desert using culture dependent method and assessment of their potential for production of industrially important enzymes. Screening of bacteria from sand dune samples collected from arid region of India led to isolation of total eight moderately thermophilic bacterial strains. All strains were characterized morphologically, biochemically and 16S rRNA gene sequencing analysis. The phylogenetic analysis of strains revealed that these strains affiliated to genera Bacillus (5 strains), Geobacillus (2 strains) and Aeribacillus (1 strain), which were able to grow between 25-60 °C. The present study documented the occurrence of species Geobacillus thermodenitificans, Bacillus paralicheniformis and Aeribacillus pallidus in sand dunes of Thar Desert, India. These moderately thermophilic strains showed an ability to produce one or more extracellular enzymes like amylase, protease, lipase and asparaginase. Thus, the strains isolated from Thar Desert could be potential candidates for industrial applications. The present study showed that bacterial strains exist in the same ecological niche might have ability to produce distinct enzymes.

Geothermal areas are the niches of a rich microbial diversity that is not only part of the intangible patrimony of a country but also the source of many microbial species with potential biotechnological applications. Particularly, microbial species in geothermal areas in Argentina have been scarcely explored regarding their possible biotechnological uses. The purpose of this work was to explore the proteolytic and keratinolytic enzymatic potential of microorganisms that inhabit in the Domuyo geothermal area in the Neuquén Province. To this end, we did enrichment cultures from two high-temperature natural samples in mineral media only supplemented with whole chicken feathers. After the isolation and the phylogenetic and morphologic characterization of different colonies, we obtained a collection of Bacillus cytotoxicus isolates, a species with no previous report of keratinolytic activity and only reported in rehydrated meals connected with food poisoning outbreaks. Its natural habitat has been unknown up to now. We characterized the proteolytic and keratinolytic capacities of the B. cytotoxicus isolates in different conditions, which proved to be remarkably high compared with those of other similar species. Thus, our work represents the first report of the isolation as well as the keratinolytic capacity characterization of strains of B. cytotixicus obtained from a natural environment.
