Antileishmanial amides and lignans from Piper cubeba and Piper retrofractum

Abstract: The n-hexane, ethyl acetate, methanol, and acetone extracts of Piper cubeba Linn. and P. retrofractum Vahl. (Piperaceae) were evaluated in vitro against promastigotes of Leishmania donovani, and all exhibited significant in vitro activity at 100 lg/ml. Two lignans, cubebin and hinokinin, were isolated from the hexane extract of P. cubeba; and one bis-epoxy lignan, (-)-sesamin, and two amides, pellitorine and piplartine, were isolated from the hexane and methanol extracts of P. retrofractum. Cubebin and piplart… Show more

“…The chemical composition of P. cubeba oil is alkaloid, amides, terpenoid, benzoic acids, furanofuran, chormenes, chalcones, lignan (cubebin, hinokinin, yatein, isoyatein), and neolignan (kadsurin and piperenone) [1]- [2], [4]- [5]. The chemicals in P. cubeba oil can be used as spices, medicines to treat many ailments (gonorrhea, dysentery, abdominal pain, diarrhea, enteritis), antileishmanial, antimicrobial, antitumoral, fungicidal, and insecticidal [1]- [3], [5]- [5]. Extraction is one of the key technologies to isolate several essential oils.…”

“…Other amides from Piper species are among the natural products of interest since many of it compounds display a broad range of antiprotozoal activities (Raay et al, 1999;Bodiwala et al, 2007;Freire-de-Lima et al, 2008). Kapil (1993) reported an investigation of piperine (amide 6) activity against promastigote forms of Leishmania donovani.…”

“…The dose and route of administration of PPTN was selected based on previous results obtained by our group. 10 The control group consisted of three rats that received water instead of PPTN solution. After administration, retro-orbital venous plexus blood samples were collected from the rats into heparinized tubes at 30 min, 3 and 24 h (three animals for each time interval).…”

“…5 In fact, PPTN, a known alkaloid amide isolated from the Piper species shows several pharmacological activities. It has been reported that this alkaloid has cytotoxic, 5,6 antifungal, 7 anti-platelet aggregation, 8 antitumoral, 5,6 anxiolytic and antidepressant, 9 antileishmanial, 10 and genotoxic 11 activities. Only one HPLC method has been described in the literature for the quantitative determination of PPTN in plants in natura, in plantlets in vitro and ex vitro, and in callus of Piper tuberculatum.…”
