Antigen requirements and specificity of a microplate enzyme-linked immunosorbent assay (ELISA) for detecting infectious bronchitis viral antibodies in chicken serum

Abstract: The conditions of a rapid, indirect-enzyme linked immunosorbent assay for Infectious Bronchitis Virus (IBV) antibodies have been established. Optimal sensitivity was obtained using 10 micrograms/ml protein concentration of the Mass 41 strain purified from infected allantoic fluid. Specificity was demonstrated with Newcastle Disease Virus (NDV) antigen-antibody system. Negligible crossreactions were observed. After bromelain or lipase treatment IBV had an ELISA reactivity similar to untreated particles suggesti… Show more

“…In poultry virology, this test has been shown to be of value to measure antibodies against infectious bronchitis virus (Mockett and Darbyshire, 1980;Soula and Moreau, 1981), reovirus (Slaght et al, 1978), infectious bursal disease virus (Marquardt et al, 1980;Howie and Thorsen, 1981), rotavirus (Bartz etal, 1980) and adenovirus (Dawson et al, 1980). The present paper describes the application of the ELISA to the detection of antibodies against infectious laryngotracheitis (ILT) virus in chicken serum.…”

“…In a different study, an enzyme immunoassay of an IBV 41 Ms serum collected 3 months post initial infection demonstrated that membrane antigens were apparently more important than surface projections, because serum reacted similarly with whole virus or the same virus treated with bromelain to remove surface projections (14). Although the observations came from experiments with two different coronaviruses, antibody to surface projections was the predominant kind early postinfection (10), and membrane antibody was the predominant kind late postinfection (14). Therefore, one explanation of the results of the present study is that the strain specificity observed after the initial exposure of IBV-susceptible broilers was due to antibody directed against the surface projections that bound only homologous antibody.…”

“…Similar results were obtained by enzyme immunoassay with antigen of purified whole virus or the surface projection subcomponent.
