Anti-inflammatory Steroid from Phragmitis rhizoma Modulates LPS-Mediated Signaling Through Inhibition of NF-κB Pathway

Abstract: Lipopolysaccharides (LPS) strongly stimulate immune cells, and unabated activation of immune system by LPS may lead to an exacerbation of sickness and depression. In this study, stigmasta-3,5-dien-7-one (ST) was isolated from Phragmitis rhizoma as a negative regulator of LPS-induced inflammation in macrophages. ST effectively reduced nitric oxide (NO), prostaglandin E2, and pro-inflammatory cytokine levels, which were markedly raised by LPS treatment. In addition, ST blocked the nuclear factor-kappa B (NF-κB) … Show more

“…Moreover, stigmasta‐3, 5‐dien‐7‐one and vanillin were probably responsible for the resistance to fungal attack. The former is known to possess anti‐inflammatory activity . The latter has since long been used in certain medical applications and as a flavoring agent, because of its inhibitory properties against fungi and bacteria and synergistic effect to enhance the antifungal effect of plant essential oils against Botrytis cinerea .…”

“…HO-1 is known as a representative antioxidant enzyme which plays an important role in protecting cells from oxidative stress and is regulated by the transcriptional activity of nuclear factor erythroid 2-related factor 2 (Nrf2) [ 53 , 54 ]. Therefore, we investigated whether phenylheptatriyne ( 1 ) expresses the antioxidant enzyme HO-1, and whether the expression of HO-1 was increased ( Figure 7 ).…”

“…Previous studies demonstrated that corticosteroids like Dex phosphate contributed to an anti-inflammatory effect through inhibiting the nuclear factor-kappa B (NF-κB) signal pathway. [5, 6] However, whether corticosteroids eye drops are proper for infectious keratitis cure remains a controversial topic. [7] Lu et al proved that dexamethone could inhibit IL-1-induced collagen degradation and decrease stroma scarring.…”
