Antagonistic effects of ?-adrenoceptor blocking agents on arrhythmias, enzyme release, and myocardial necrosis in isolated rat hearts with coronary occlusion and reperfusion

Abstract: In isolated perfused rat hearts reperfusion of the occluded left coronary artery led to arrhythmias, their severity depending on the duration of the foregoing period of myocardial ischaemia. Simultaneously, high activities of the myocardial enzyme creatine kinase (CK) were released into the perfusion fluid. Corynanthine, blocking mainly alpha 1-adrenoceptors, and rauwolscine, blocking mainly alpha 2-adrenoceptors, concentration-dependently antagonized the reperfusion-induced arrhythmias (3-30 mumol/l). The mos… Show more

“…Therefore, the occurrence of cardiac arrhythmias for up to 3 min was assigned the factor 2; 3-6 min was assigned the factor 4; 6 -10 min was assigned the factor 6; 10 -15 min was assigned the factor 8; 15-20 min was assigned the factor 10; 20 -25 min was assigned the factor 11; and 25-30 min was assigned the factor 12. Values ranging from 2 to 12 were obtained and were denoted as the arrhythmia severity index (ASI) (1,9).…”

“…A value of 0 to 12 was thus obtained in each experiment and is denoted as arrhythmia severity index (ASI). 12 To evaluate the role of cyclooxygenase products in the Ang-(1-7) effects, rats received indomethacin (5 mg/kg IP) plus heparin (400 IU IP). After 1 hour, the rats were decapitated, the thorax was opened, and the heart was dissected and perfused with KRS or KRS containing Ang-(1-7) (0.22 nmol/L, nϭ8).…”

“…Therefore, the occurrence of cardiac arrhythmias for up to 5 min was assigned the factor 1, 10 min was assigned the factor 2, 15 min was assigned the factor 3, 20 min was assigned the factor 6, 25 min was assigned the factor 9, and 30 min was assigned the factor 12. A value of 0-12 was thus obtained in each experiment and is denoted "arrhythmia severity index" (ASI) (16,17). Data are reported as means ± SEM.…”
