Abolition of reperfusion-induced arrhythmias in hearts from thiamine-deficient rats

Abstract: tensive work has been done regarding the impact of thiamine deprivation on the nervous system. In cardiac tissue, chronic thiamine deficiency is described to cause changes in the myocardium that can be associated with arrhythmias. However, compared with the brain, very little is known about the effects of thiamine deficiency on the heart. Thus this study was undertaken to explore whether thiamine deprivation has a role in cardiac arrhythmogenesis. We examined hearts isolated from thiamine-deprived and control … Show more

“…The confocal line-scan imaging was performed with a Zeiss LSM 510META confocal microscope. The amplitude of the Ca 2ϩ transient evoked by the application of a Ca 2ϩ -and Na ϩ -free solution containing 10 mM caffeine was used as an indicator of the SR Ca 2ϩ load (36). Cells were subjected to a series of preconditioning pulses (1 Hz) before caffeine was applied.…”

“…An EPC-9.2 instrument (HEKA Electronics) was used to patch clamp single ventricular cardiomyocytes in whole-cell voltage and current clamp configurations, using specific protocols according to the ionic conductance evaluated (8,36,42). Measurements started 5 min after breaking into the cell in order to attain equilibrium between the pipette solution and the cell cytoplasm.…”

“…Whole-cell voltage-clamp recordings were obtained at room temperature (22-25 • C) using an EPC-9.2 patch clamp amplifier (HEKA Electronics, Rheinland-Pfalz, Germany) as has been previously described Oliveira et al, 2007;Roman-Campos et al, 2009;Lara et al, 2010). For I Ca,L measurements, recording pipettes were filled with internal solution containing (in mM) 5 NaCl, 120 CsCl, 20 TEA-Cl, 5 EGTA, and 10 HEPES (pH set to 7.2 with CsOH).…”

“…On the other hand, the major manifestations of wet beriberi are tachycardia, cardiomegaly, high-output heart failure, and pedal edema, in addition to neuropathy. Direct impairment of myocardial energy production has been proposed as a possible basis for the development of heart failure state seen in beriberi (because of loss of thiamine's beneficial role in metabolic reactions) [6]. Decreased activity of pyruvate dehydrogenase due to thiamine deficiency results in build-up of pyruvate, which is then shunted towards anaerobic conversion into lactate [7].…”
