Androgen Receptor-Related Non-coding RNAs in Prostate Cancer

Abstract: Prostate cancer (PCa) is the second leading cause of cancer-related death among men in the United States. Androgen receptor (AR) signaling is the dominant oncogenic pathway in PCa and the main strategy of PCa treatment is to control the AR activity. A large number of patients acquire resistance to Androgen deprivation therapy (ADT) due to AR aberrant activation, resulting in castration-resistant prostate cancer (CRPC). Understanding the molecular mechanisms underlying AR signaling in the PCa is critical to ide… Show more

“…Competing roles of miRNAs and lncRNAs in regulating gene expression and an additional role of miRNAs in repressing translation in AR-independent prostate cancer and in NEPC have been reported [ 88 ]. There is also a direct impact of a number of non-coding RNAs on AR gene transcription, whereas the level of several of them is controlled by the AR [ 89 , 90 , 91 ]. Shifts in non-coding RNA abundance following therapy and an implication in resistance to AR antagonists or taxanes have been reported [ 92 ].…”

“…An alternative mechanism for lncRNAs to influence the AR is through interactions with miRNAs. Although the contribution of numerous miRNAs and lncRNA in CRPC development has been summarized and reviewed in the literature, these authors only elaborated on the impact of these two ncRNAs separately without sorting out the interaction of lncRNAs and miRNAs [ 47 , 48 ]. There is accumulating evidence proposed that cancer development is highly correlated with the interaction patterns between lncRNAs and miRNAs [ 49 , 50 ].…”

“…Long noncoding RNA (lncRNA), especially androgen receptor-related lncRNA, had the importantly clinical utility in the tumor progression, diagnosis and prognosis, and as therapeutic targets of prostate cancer 6 8 . LncRNA NEAT1 promoted bone metastasis via miR-205-5p in prostate cancer 9 .…”
