In recent years, oxidative stress has been postulated to be an important factor in the pathogenesis and development of lifestyle-related diseases. In this study, we investigated the association between the derivatives of reactive oxygen metabolites (d-ROMs), as an index of products of reactive oxygen species (ROS), and biological antioxidant potential (BAP), as an index of antioxidant potential. We also investigated the associations between d-ROMs or BAP and the risk factors for lifestyle-related diseases or metabolic syndrome-associated factors to evaluate their usefulness in preventive medicine. There were 442 subjects who underwent health checkup examination in our facilities. In addition to standard medical checkup items, we analyzed d-ROMs, BAP, brachial-ankle pulse wave velocity, high-sensitivity C-reactive protein level and visceral fat area (VFA) visualized on a computed tomography scan. The mean d-ROM value in females was significantly higher than that in males. There was a positive correlation between the d-ROM and VFA levels. On correlation analysis, there was a negative correlation between the d-ROM and creatinine levels. As factors that influence d-ROMs, the level of VFA was selected, suggesting the significance of oxidative stress measurement with d-ROMs. In addition, there was a positive correlation between d-ROMs and BAP values. Further research is required to resolve whether increased production of ROS or the antioxidant potential that can compensate for such an increase of ROS is more important in vivo. INTRODUCTIONOxidative stress may be involved in various disorders and pathogeneses, especially in lifestyle-related diseases, and it causes the progression and acceleration of atherosclerosis. 1-3 Biomarkers of oxidative stress, such as serum malondialdehyde level measured as thiobarbituric acid-reactive substances, oxidized low-density lipoprotein, oxidation injury of DNA, 8-hydroxydeoxyguanosine and urinary excretion of 8-iso-prostaglandin-F2a, are generally measured in laboratories. [4][5][6][7] On the other hand, as indices of antioxidant potential, intracellular levels of superoxide dismutase and glutathione peroxidase have mainly been measured. [8][9] However, these assay methods are complex and are not suitable for analysis of a large number of subjects. Recently, simpler methods for detecting reactive oxygen species (ROS) using derivatives of reactive oxygen metabolites (d-ROMs) and biological antioxidant potential (BAP) were developed, and reports on studies using these methods have been increasing. [10][11][12][13][14][15][16][17][18][19][20] Thus, in the present study, we measured d-ROMS and BAP as indices for the production of ROS and antioxidant potential, respectively.

This study aimed to investigate antioxidant and anti-inflammatory activities of a commercial product of noni (Morinda citrifolia) juice. Carrageenan-induced rat paw edema was employed as inflammatory model. One control and three experimental groups were formed. Experimental groups were administered noni juice alone, noni juice+carrageenan, and carrageenan alone. Oxidant and antioxidant capacity were determined by d-ROMs test and BAP test, respectively. Plasma concentrations of endothelin-1 and leptin were measured by ELISA. Measurements were performed at zero time and 2nd hour of inflammation. Oxidant capacity decreased in noni-received groups at 2nd hour (p=0.019). Antioxidant capacity of the group which received noni alone was found to be higher at 2nd hour (p=0.036). Plasma concentrations of endothelin-1 and leptin were notably lower in noni-received groups (p=0.001 and p=0.021, respectively). The results show that the commercial noni juice investigated has pronounced antioxidant and anti-inflammatory activities.

Background The aims of this study were (a) to ascertain age‐related changes in the reference values in hematological and serum biochemical examinations of beagles, and (b) to clarify the changes in these findings, including acute phase proteins and oxidative stress, throughout pregnancy and after parturition. Methods Clinicopathological parameters were measured in young beagles at 6, 9 and 12 months and in adult beagles aged from 24 to 60 months. Likewise, pregnant beagles were investigated throughout the pregnancy and after parturition. Results Apparent age‐related changes were found in erythrocytic parameters during the growth and development of beagles. Most of the parameters (total protein, albumin, blood urea nitrogen, creatinine, urate, alkaline phosphatase (ALP) and creatine kinase (CK) exhibited age‐dependent transitions. White cell count significantly increased after 30 days of pregnancy. The values of erythrocytic parameters moderately decreased during the second half of the pregnancy. Triglycerides, total cholesterol, free cholesterol and phospholipid concentrations increased in the mid‐ and late stages of pregnancy. ALP, lactate dehydrogenase, CK and cholinesterase activities markedly increased during pregnancy and/or after parturition. C‐reactive protein (CRP) concentrations gradually increased and reached a maximum after 30‐40 days of pregnancy. Serum amyloid A (SAA) levels markedly increased at 30 days of pregnancy before subsiding, and then increased again 3 days after parturition. Reactive oxygen metabolites (d‐ROMs) showed significant increases after 30 and 40 days of pregnancy. Conclusions Reference values for hematological and serum biochemical examinations should be used for health evaluation of dogs, taking sex, age and the stage of pregnancy into consideration. Measurements of CRP, SAA and d‐ROM levels are also useful for assessing maternal conditions in mid‐pregnancy.
