Anaerobiosis in Echinochloa crus-galli (Barnyard Grass) Seedlings

Abstract: Echinochloa crus-galli var oryzicola (hereafter oryzicola) is a common weed of flooded rice fields. Like rice (4, 7) oryzicola can germinate and grow for long periods without 2 (10). In the past, studies of anaerobic metabolism have concentrated mainly on the effects of flooding on plant roots with little attention directed toward germinating seeds (7). This is despite the evidence that most flood-tolerant plant roots are merely surviving temporary stress (4); whereas tolerant seeds, such as oryzicola and rice… Show more

“…By internal amino acid sequence analysis, we have preliminarily identified this polypeptide as enolase (EC 4.2.1.11) (unpublished data). The significance of this finding relative to our previous biochemical studies on the anaerobic metabolism of E. phyllopogon (Rumpho and Kennedy, 1981, 1983a, 1983b) is being pursued.…”

“…Vol. 101, 19931983b, glycolysis (Rumpho and Kennedy, 1983a;Kennedy et al, 1987b), the oxidative pentose phosphate pathway (Kennedy et al, 1983;Rumpho and Kennedy, 1983a), lipid metabolism (VanderZee and Kennedy, 1982;Knowles and Kennedy, 1984;Everard and Kennedy, 1985), and mitochondrial activity (Kennedy et al, 1987a;Fox and Kennedy, 1991) operate under anaerobic conditions. Recently, we (Fox and Kennedy, 1991) reported that a complete TCA cycle was present in E. phyllopogon under anoxia and that most TCA cycle enzymes in nitrogen-grown seedlings exhibited in vitro activities comparable to those grown in air.…”

“…var oryzicola (hereafter, crusgalli and oryzicola, respectively), rice (Oryza sativa L., cv S201) pea (Pisum sativum L., cv Alaska), and wheat (Triticum aestivum L., cv Dawes) were surface-sterilized in 2.5% NaOCl for 10 min, washed three times with glass-distilled H20, and germinated with the appropriate N2 (99.995% N2) or air gas phase as before (13). No bacterial or fungal contamination was detected when an aliquot of the germination solution was incubated on agar plates containing (per liter): 15 g agar, 3 g yeast extract, and 10 g glucose (20). Data Figure 2.…”

“…Recently, we reported (13) In previous reports, we have presented information on the structural aspects of anaerobic germination in Echinochloa seeds (22), seedling characteristics and cellular ultrastructure (13), chloroplast ultrastructure (23), and metabolism of anaerobic germination (18)(19)(20). This paper presents information on the respiration and response to respiratory inhibitors of anaerobically grown Echinochloa seedlings.…”
