An oligonucleotide microarray for microRNA expression analysis based on labeling RNA with quantum dot and nanogold probe

Abstract: MicroRNAs (miRNAs) play important regulatory roles in animals and plants by targeting mRNAs for cleavage or translational repression. They have diverse expression patterns and might regulate various developmental and physiological processes. Profiling miRNA expression is very helpful for studying biological functions of miRNAs. We report a novel miRNA profiling microarray, in which miRNAs were directly labeled at the 3′ terminus with biotin and hybridized with complementary oligo-DNA probes immobilized on glas… Show more

“…miRNAspecific quantitive PCR and microarray technology make this study become easier. In the past 1 year, several quantitive PCR Raymond et al, 2005;Shi and Chiang, 2005) and microarray technology (Babak et al, 2004;Barad et al, 2004;Nelson et al, 2004b;Thomson et al, 2004;Liang et al, 2005) have been developed, and successfully used to study the functions of miRNAs in organism development (Miska et al, 2004), disease and cancer (Lu et al, 2005b). At present, several companies have developed miRNA chips for studying the expression profiles at a specific situation.…”

“…Knockdown and/or knockout or overexpression of specific miRNAs is a good approach to investigate the specific function of an unique miRNA (Hutvagner et al, 2004;Meister et al, 2004a;Lee et al, 2005). Recently developed miRNA microarray technology and miRNAspecific real-time PCR also provide useful information on miRNA functions (Miska et al, 2004;Baskerville and Bartel, 2005;Liang et al, 2005).…”

“…Several different approaches enabling multiplexed detection of miRNAs using microarray technologies have been reported (Krichevsky et al, 2003;Babak et al, 2004;Liu et al, 2004;Miska et al, 2004;Nelson et al, 2004;Sun et al, 2004;Thomson et al, 2004;Liang et al, 2005), including one from our laboratory (Baskerville and Bartel, 2005). In our approach, probes consist of T m -normalized DNA oligonucleotides antisense to the given small RNA sequence.…”

“…It has been assumed that most, if not all, biosensing measurements rely on the information of the final complex followed by host-guest molecules association. Nevertheless, several binding events cannot form a stable complex, and the binding process is dynamic, even reversible, with a weak binding strength, which poses a great challenge for routine detection tools, like microarray that involves tedious fluorescent labeling and is generally incapable of real-time monitoring (Liang et al 2005, Li et al 2010). Thus, a reliable, label-free, and real-time transduction strategy for molecular interactions is highly desirable.…”
