Black Mulberry (Morus nigra l.) belongs to Moraceae family. The present study evaluated the possible genotoxic and/or protective activities of black mulberry fruit juice (BMFJ), in vitro, using mitomycin C (MMC) as a positive control, by chromosomal aberrations and micronucleus assays. Human lymphocytes were treated with BMFJ concentrations alone (1/1, 1/2, 1/4, 1/8 dilutions), pretreatment (49h) (0.20 μg/ml MMC+ 1/1 BMFJ, 0.20 μg/ml MMC+1/2 diluted BMFJ, 0.20 μg/ml MMC+1/4 diluted BMFJ, 0.20 μg/ml MMC+1/8 diluted BMFJ) and simultaneous-treatment (48h) (0.20 μg/ml MMC+ 1/1 BMFJ, 0.20 μg/ml MMC+1/2 diluted BMFJ, 0.20 μg/ml MMC+1/4 diluted BMFJ, 0.20 μg/ml MMC+1/8 diluted BMFJ). The in vitro results demonstrated that BMFJ showed no genotoxicity, but it significantly decreased chromosomal aberration and micronucleus frequency induced by MMC. Our results showed that all concentrations of BMFJ revealed no genotoxicity but protective activity against genomic changes induced by anti-tumor agent MMC in human lymphocytes. Protective effects of BMFJ on MMC induced chromosomal damages most probably due to its free radical scavenging activity.

One of the promising directions in the fight against the emergence and spread of farm animal microbiota resistance factors is the development and search for feed additives that can inhibit the SOSresponse. SOS-response is one of the main mechanisms of the occurrence of mutations in bacteria. Plants used in traditional medicine can be a promising source of safe substances that reduce the SOS-response of bacteria. A screening of plants potentially containing substances with antiSOS activity was performed. During the initial screening, the E. coli MG 1655 pRecA-lux biosensor strain with ciprofloxacin as RecA inducer was used. Seven plants were identified whose extracts reduced the expression of the RecA operon. In further experiments on bacteria exposed to antibiotics, we identified four plants whose exstracts significantly reduced the mutagenesis rate of clinical E. coli strains: Austrian broom (Cytisus austriacus), greater celandine (Chelidonium majus), walnut (Juglans regia) and smooth sumac (Rhus glabra).

Aqueous extracts were prepared from eight medicinal plants and other plants were prepared as essential oils.The radical-scavenging ability of each plant extract was determined by the 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging assay. The total phenolic content of plants was determined by the Folin-Ciocalteu reagent in terms of gallic acid equivalents. The DPPH scavenging potential of the aqueous extracts ranged from 17 to 79%, whereas the essential oils showed inhibition of the DPPH activity in the 12-88% range. The highest inhibition of DPPH radicals was observed for Pinus halepensis extract. Meanwhile, amongst the essential oils, the greatest antioxidant potential was exhibited by Nigella sativa. The Bauhinia variegate extract had the highest phenolic content (149.18 mg/g gallic acid equivalents), followed by Albizzia lebbeck (148.00 mg/g) and Pinus halepensis (145.67 mg/g), whereas, amongst the essential oils, the highest phenolic content (98.57 mg/g) was found for Thymus vulgaris. The lowest contents were observed for Kigelia africana and Rosmarinus officinalis.The antioxidant activity had a positive correlation (R=0.654) with the phenolic content of most aqueous extracts, whereas it had a weak correlation using the essential oils (R=0.335). This confirms that the phenolic content of aqueous extracts may contribute towards their antioxidant properties.
