An efficient regeneration system via direct and indirect somatic embryogenesis for the medicinal tree Murraya koenigii

Abstract: A reproducible protocol for direct and indirect somatic embryogenesis was established in a small aromatic tree, Murraya koenigii. Embryogenic callus was obtained from 90% zygotic embryonic axis (ZE) and 70% cotyledon (COT) explants in Murashige and Skoog (MS) basal medium supplemented with 8.88 lM 6-benzyladenine (BA) and 2.675 lM a-naphthaleneacetic acid (NAA). Globular somatic embryos were induced and further matured from such embryogenic callus by subsequent culture on the same basal media containing thidia… Show more

“…TDZ has been proved to be a potential PGR and acts as a substitute for both the auxin and cytokinin requirements (Murthy et al, 1998). Plant regeneration by using TDZ was reported in several plants like Murraya koenigii (Paul et al, 2011) Jatropha curcas (Kumar and Reddy, 2012), Stevia rebaudiana (Lata et al, 2013). In some reports, TDZ was found to facilitate the shoot elongation and root induction (Debnath, 2005) by promoting regulated plant morphogenesis through the modulation of endogenous cytokinin and auxin (Gill and Saxena, 1992;Thomas and Katterman, 1986).…”

“…We have achieved 86% direct regeneration in EIM2 and obtained apparently normal plants in a short period of culture. Both factors have been described as convenient, to reduce the occurrence of somaclonal variation (Cote et al, 2001;Sharma et al, 2007;Paul et al, 2011). This is the first report that describes the use of mature seeds as explants for embryogenesis induction in grape.…”

“…In several plant species somatic embryogenesis is induced in the presence of auxin alone or in combination with cytokinins 24,25,26,27 and in a few systems cytokinins alone were found to be effective in somatic embryogenesis 28,29 . In this study it was found that the young shoot buds induced direct somatic embryogenesis at high BAP concentration (5 mg L-1 ) while its lower conc.…”
