Background: Cadmium (Cd) is a severely toxic heavy metal to most microorganisms. Many bacteria have developed Cd 2+ resistance. Results: In this study, we isolated two different Cd 2+ resistance Bacillus sp. strains, Bacillus vietamensis 151-6 and Bacillus marisflavi 151-25, which could be grown in the presence of Cd 2+ at concentration up to 0.3 mM and 0.8 mM, respectively. According to the genomic sequencing, transcriptome analysis under cadmium stress, and other related experiments, a gene cluster in plasmid p25 was found to be a major contributor to Cd 2+ resistance in B. marisflavi 151-25. The cluster in p25 contained orf4802 and orf4803 which encodes an ATPase transporter and a transcriptional regulator protein, respectively. Although 151-6 has much lower Cd 2+ resistance than 151-25, they contained similar gene cluster, but in different locations. A gene cluster on the chromosome containing orf4111, orf4112 and orf4113, which encodes an ATPase transporter, a cadmium efflux system accessory protein and a cadmium resistance protein, respectively, was found to play a major role on the Cd 2+ resistance for B. vietamensis 151-6. Conclusions: This work described cadmium resistance mechanisms in newly isolated Bacillus vietamensis 151-6 and Bacillus marisflavi 151-25. Based on homologies to the cad system (CadA-CadC) in Staphylococcus aureus and analysis of transcriptome under Cd 2+ induction, we inferred that the mechanisms of cadmium resistance in B. marisflavi 151-25 was as same as the cad system in S. aureus. Although Bacillus vietamensis 151-6 also had the similar gene cluster to B. marisflavi 151-25 and S. aureus, its transcriptional regulatory mechanism of cadmium resistance was not same. This study explored the cadmium resistance mechanism for B. vietamensis 151-6 and B. marisflavi 151-25 and has expanded our understanding of the biological effects of cadmium.

Sensitive seedling crops have been developed to monitor Cadmium (Cd) contamination in agricultural soil. In the present study, 18 parameters involving growth conditions and physiological performances were assessed to evaluate Cd-responses of three wheat (Triticum aestivum L.) cultivars, Xihan1 (XH), Longzhong1 (LZ) and Dingfeng16 (DF). Principle component analysis illustrated that Factor 1, representing growth performance, soluble sugar content and catalase activity, responded to the Cd treatments in a dose dependent manner, while Factor 2 represented by chlorophyll content and germinating root growth was mainly dependent on cultivar differences. Higher inhibition rates were observed in growth performance than in physiological responses, with the highest inhibition rates of shoot biomasses (39.6%), root length (58.7%), root tip number (57.8%) and bifurcation number (83.2%), even under the lowest Cd treatment (2.5 mg·L−1). According to the Cd toxicity sensitivity evaluation, DF exerted highest tolerance to Cd stress in root growth while LZ was more sensitive to Cd stress, suggesting LZ as an ideal Cd contaminant biomarker. This study will provide novel insight into the cultivar-dependent response during using wheat seedlings as Cd biomarkers.

Cadmium (Cd) is a non-essential and harmful element to humans. Cadmium contamination is a serious issue for human health, especially in densely populated agroecology areas. In this study, the investigation of an agroecology area was conducted to gain insight into the relationship between Cd in wheat and soil and then evaluate the Cd total risk for human health. The soil samples and their matching wheat samples, underground water samples, and atmospheric deposition (air) samples were collected from a wheat-growing area in an agroecology plain. The cadmium concentration in the four types of media, in order, was air > soil > wheat > water. The mean concentration of the geoaccumulation index (Igeo) showed that the total Cd in soil (Cd-T) and Cdair reached a mild and moderate pollution level. The results of the correlation and principal component analysis (PCA) showed that the majority of Cdwheat originated from Cd-2 (exchangeable), Cd-4 (humic acid-bound), and Cd-7 (residual). Furthermore, the results of the stepwise multiple linear regression (SMLR) showed that three fractions were primarily controlled by Cd-T: clay, cation exchange capacity (CeC), and total organic carbon (TOC). In addition, the total cancer risk (CR) of Cd in multiple media was, in the order wheat > water > soil > air. It is noteworthy that the Cd content in underground water and wheat by the ingestion pathway posed cancer risks to the local residents and provided a comprehensive insight into multiple media environment management. Furthermore, it provides a very significant basic study for detailed research into the mobility and transformation for factions.
