Air Pollutant Sulfur Dioxide‐induced Alterations on the Levels of Lipids, Lipid Peroxidation and Lipase Activity in Various Regions of the Rat Brain

Low-density lipoprotein oxidation is implicated in the development of atherosclerosis. Plasma susceptibility to oxidation may be used as a marker of low-density lipoprotein oxidation and thus predict atherosclerotic risk. In this study the authors investigated the relationship between plasma susceptibility to oxidation and exposure to automotive pollution in a group of automobile mechanics (n = 16) exposed to high levels of automotive pollution, vs. matched controls (n = 13). The authors induced plasma oxidation by a free radical initiator and they determined susceptibility to oxidation by (1) change in absorbance at 234 nm, (2) lag time to conjugated diene formation, and (3) linear slope of the oxidation curve. Mechanics had significantly higher values (mean +/- standard error) for change in absorbance (1.60 +/- 0.05 vs. 1.36 +/- 0.05; p < .002), and slope (1.6 x 10(-3) +/- 0.1 x 10(-3) vs. 1.3 x 10(-3) +/- 0.1 x 10(-3); p < .001), compared with controls. These results indicate that regular exposure to automotive pollutants increases plasma susceptibility to oxidation and may, in the long-term, increase the risk of developing atherosclerosis.

The effects of vitamin E (alpha-tocopherol) on the rate of lipid peroxidation and lipase activity in the discrete areas of the brain and spinal cord were studied. Administration of metasystox in two different doses 1.0 and 2.0 mg/kg body weight intraperitoneally daily for 10 days have shown a dose-related depletion of total lipids, cholesterol and esterified fatty acids in the rat cerebral hemisphere, cerebellum, brain stem and spinal cord. The rate of lipid peroxidation and lipase activity showed dose-dependent increase in different regions of the CNS. Administration of alpha-tocopherol alone caused significant reduction of lipid peroxidation and lipase activity. When it was administered simultaneously with metasystox, alpha-tocopherol prevented the metasystox-induced increase of lipid peroxidation and lipase activity.

The effects of the pesticide, metasystox (0,O-dimethyl-S-2 or MetasystorB (ethylsulphinyl) ethylthiophosphate), on various lipid fractions in the discrete areas of the brain were studied. The daily intraperitoneal administration of 4 mg/kg body weight of metasystox for 10 days has depleted the levels of total lipids, phospholipids, cholesterol, esterified fatty acids and gangliosides in cerebral hemisphere, cerebellum, brain stem and spinal cord. But the lipase and lipid peroxidation measured in the cerebral hemisphere were significantly increased. It is possible that the inhibition of the lipid levels in the discrete areas of the brain may be due to the increase activity of lipase and lipid peroxidation.
