Nile red is a widely used hydrophobic dye for probing the structure, dynamics, and environment in many biological and microheterogeneous systems. This paper reports emission-wavelength-dependent fluorescence intensity decay of Nile red in membranes and micelles. Global analysis of these multiple fluorescence decays reveals a double-exponential decay with negative amplitudes for the short-lifetime component at longer emission wavelengths. This indicates an excited-state kinetics leading to the formation of a new species in the excited state from the initially excited state. In both the cases, the short lifetime corresponds to that of the initially excited species. This excited-state kinetics is also observed in the case of viscous organic solvents such as 1-octanol and glycerol and is attributed to that of an excited-state solvent relaxation.

The spectroscopy of nile red (NR) was reported when incorporated in organically modified silicates (ORMOSILs), obtained by the copolymerization of (glycidoxypropyl)trimethoxysilane and tetramethoxysilane in various proportions. An increase of the fluorescence intensity, a gradual blue spectral shift by 43 nm in absorption and 45 nm in emission, and a change in the lifetime from 2.6 to 4.4 ns of the dye were observed as a response to the change of the polarity and the structure of the ORMOSILs caused by the addition of (glycidoxypropyl)trimethoxysilane. The decrease of the nonradiative H + -NR form was observed as one of the consequence of addition of (glycidoxypropyl)trimethoxysilane. Absorption spectral shifts of the NR were used for the determination of a polarity empirical scale (E NR ) of the ORMOSIL. The value of E NR (r 0/100 ) ) 48.70 kcal/mol for NR in silica xerogels prepared from tetramethoxysilane and E NR (r 100/0 ) ) 52.55 kcal/mol for NR in ORMOSILs prepared from (glycidoxypropyl)trimethoxysilane showed the high polarity of the inorganic matrixes in contrast to ORMOSILs. A study in solution was performed in order to make a concluding comparison of the NR behavior in terms of the restricted environment around the NR molecule ORMOSILs.

The global health scenario in present times has raised human awareness about drug delivery strategies. Among colloidal drug delivery vehicles, vesicular nanocarriers such as liposomes and niosomes are popular. However, liposomes and niosomes get disrupted in the harsh environment of the gastrointestinal tract. In this context, the drug delivery community has reported the superior performance of vesicles containing bile salts, that is, bilosomes. The present work attempts to examine the structural/morphological aspects underlying the superior performance of bilosomes. Optical microscopy, electron microscopy, and light scattering give a definite proof of the enhanced stability of bilosomes compared to niosomes, both prepared from the same amphiphilic molecule. Fluorescence probing of the vesicles provides detailed insight into the bilayer characteristics and the differences between bilosomes and niosomes. Fluorescence resonance energy transfer studies lend further support to the findings that bilosomes have a more flexible bilayer structure than niosomes. The entrapment efficiency of the vesicles for the well-known antioxidant curcumin (whose bioavailability is a matter of concern due to low water solubility) was also studied. Bilosomes show higher curcumin entrapment efficiency than niosomes. For use in drug delivery, one needs to establish a trade-off between cargo/drug entrapment and release. Thus, a flexible bilayer structure is an advantage.
