Aflatoxin B1Induced Compositional Changes in Gut Microbial Communities of Male F344 Rats

Abstract: Aflatoxins are a group of potent foodborne toxicants naturally occurring in maize and groundnuts. Differential species-specific sensitivity to aflatoxins has been documented but cannot be fully explained by the differences in metabolism of these toxicants among animal species. Commensal microbial communities (microbiota) are critical to human and animal health, but few studies have assessed interactions between xenobiotic toxins and those microbiota, and its potential effects to humans and animals. Here, an ex… Show more

“…Extracted DNA was amplified in replicates using two rounds of PCR, using the forward: S-D-Bact-0341-b-S-17 (5 0 -CCTACGGGNGGCWGCAG-3 0 ); and reverse: S-D-Bact-0785-a-A-21 (5 0 -GAC TACHVGGGTATCTAATCC-3 0 ) 16S rRNA primer pair [13]. These primers were modified with the addition of Illumina TruSeq sequences and unique 8 + 12, forward and reverse, variable length barcodes [14,15]. Positive amplification was verified on a 1.5% agarose gel and equal amounts of each sample were pooled and cleaned with SPRI-beads (Thermo Fisher Scientific, Waltham, MA) with a 0.92:1 ratio.…”

“…It is also plausible that bacteria can adhere to enterocytes in the small intestine and then generate a biofilm that protects against AFB 1 contact with local cells. In addition, some bacteria can produce bactericides that also might facilitate AFB 1 elimination (Wang et al 2015). Clearly, the exact mechanisms remain to be determined to explain how the LP and MT, moreover in concert, are acting in situ to mitigate AFB 1 toxicity.…”

“…With dysbiosis, binding and absorption of mycotoxin is enhanced. 125,126 This observation is bolstered by the finding that probiotic supplementation significantly reduces the biologically available effective toxic dose of aflatoxin. With their ability of normalizing gut microbiota, probiotics potentially offer an effective dietary approach to reduction of risk for liver cancer.…”
