The Lycopersicon pennellii accession LA716 has been used as a source of arthropod pest resistance in tomato breeding programs due the high contents of acylsugars presents in its leaflets. We investigated the relationship between high foliar acylsugar contents and repellence to spider mite Tetranychus evansi in plants with contrasting acylsugar levels, selected from F 2 and BC 1 F 2 (= F 2 of the first backcross towards L. esculentum) generations derived from the interespecific cross L. esculentum 'TOM-584' × L. pennellii LA716. Mite resistance was assessed by a repellence test. Plants selected for high levels of acylsugars in leaflets had mite repellence levels similar to that of LA716. The high correlation confirmed the association between high acylsugar levels and mite repellence. Foi verificada a correlação entre o teor de acilaçúcares em folíolos de tomateiros e a repelência ao ácaro Tetranychus evansi Baker & Pritchard. Foram selecionadas plantas contrastantes quanto aos níveis de acilaçúcares nos folíolos de populações F 2 e F 2 RC 1 (= F 2 do primeiro retrocruzamento para Lycopersicon esculentum) do cruzamento interespecífico de L. esculentum TOM 584 (baixo teor de acilaçúcares) e L. pennellii LA 716 (alto teor). Foi realizado um teste de repelência ao ácaro T. evansi, tomando por base a distância percorrida pelos ácaros nos folíolos. Os genótipos selecionados para alto teor de acilaçúcares, em média, foram responsáveis por reduções significativas nas distâncias percorridas pelos ácaros sobre a superfície do folíolo. A repelência ao ácaro T. evansi exercida pelas plantas selecionadas para alto teor de acilaçúcares deu-se de forma semelhante àquela conferida pelo genitor LA-716. As magnitudes das correlações encontradas confirmam a associação entre altos teores do aleloquímico e a repelência ao ácaro. ResumoCom o presente trabalho, objetivou-se estimar a capacidade combinatória de linhagens de tomate ricas em acilaçúcares (AA) quando cruzadas com linhagens com baixo teor de acilaçúcares e estudar a expressão da heterose nos híbridos obtidos. O experimento foi realizado no campo da HortiAgro Sementes Ltda, localizada em Ijaci-MG. O material genético foi constituído de seis híbridos experimentais em esquema de dialelo parcial, constituído de três genitores masculinos (grupo I, com altos teores de acilaçúcares) e dois genitores femininos (grupo II, com baixos teores). Foram avaliadas as características: massa média de fruto (g fruto -1 ) e produção por hectare (t ha -1 ). As linhagens TOM-687 e TOM-688 (ricas em AA e resistentes a pragas) apresentaram estimativas positivas dos efeitos de capacidade geral de combinação, para massa média de frutos, e estimativas negativas, para produção, o que significa que elas contribuíram positivamente para aumento no tamanho do fruto, mas reduziram a produtividade dos híbridos. Já a linhagem TOM-689, ao contrário das duas anteriores, contribuiu para reduzir o tamanho do fruto, mas contribuiu para aumentar a produtividade dos híbridos. Os híbridos obtidos do cruzamento de linhagens com altos teores de acilaçúcares mostraram-se competitivas quando comparadas a híbridos comerciais encontrados no mercado. As melhores combinações híbridos foram obtidas quando as linhas com altos teores de acilaçúcares foram cruzadas com a linhagem elite TOM-684. AbstractThe objective of this work was to assess heterosis and combining ability of tomato lines with high acylsugar (AS) content (group I) and lines with low AS (group II) used to produce AS-rich tomato hybrids. The trial was carried out at the seed production station of the HortiAgro Sementes Ltda., at Ijaci-MG. The genetic material consisted of six experimental hybrids obtained from crosses between three male parents with high AS (group I) and two female parents with low AS (group II). Average fruit weight (g fruit -1 ) and total yield (t ha -1 ) were the traits assessed. The AS-rich pest-resistant lines TOM-687 and TOM-688 showed positive estimates of general combining ability for average fruit mass and negative for yield, increasing fruit size, and reducing the yield of the hybrids. Unlike the two previous lines, TOM-689 presented negative estimates for fruit mass and positive for fruit yield. The data showed that it is feasible to get competitive hybrids from lines with AS sugar content. The best hybrids were produced when these lines were crossed with TOM-684.

Plant glandular secreting trichomes are epidermal protuberances that produce structurally diverse specialized metabolites, including medically important compounds. Trichomes of many plants in the nightshade family (Solanaceae) produce O-acylsugars, and in cultivated and wild tomatoes these are mixtures of aliphatic esters of sucrose and glucose of varying structures and quantities documented to contribute to insect defense. We characterized the first two enzymes of acylsucrose biosynthesis in the cultivated tomato Solanum lycopersicum. These are type I/IV trichome-expressed BAHD acyltransferases encoded by Solyc12g006330─or S. lycopersicum acylsucrose acyltransferase 1 (Sl-ASAT1)─and Solyc04g012020 (Sl-ASAT2). These enzymes were used-in concert with two previously identified BAHD acyltransferases-to reconstruct the entire cultivated tomato acylsucrose biosynthetic pathway in vitro using sucrose and acyl-CoA substrates.Comparative genomics and biochemical analysis of ASAT enzymes were combined with in vitro mutagenesis to identify amino acids that influence CoA ester substrate specificity and contribute to differences in types of acylsucroses that accumulate in cultivated and wild tomato species. This work demonstrates the feasibility of the metabolic engineering of these insecticidal metabolites in plants and microbes.Solanum | glandular trichomes | acylsugar | specialized metabolism | genotype to phenotype P lants are masters of metabolism, producing hundreds of thousands of small molecules known as specialized metabolites, which vary widely in structure, abundance, and physical and biological properties. These metabolites tend to be produced by enzymes that evolve faster than those that produce "central" metabolites such as amino acids, nucleotides, sugars, and cofactors (1-3), and the pathways and metabolic intermediates involved in biosynthesis of many specialized metabolites remain mysterious. Despite the growing availability of genomic DNA sequences, understanding the genetic and biochemical mechanisms that contribute to this phenotypic diversity and plasticity presents enduring and major challenges in plant biochemistry. It is of great interest to understand and manipulate the biosynthesis of these biologically active molecules.Specialized metabolites typically are produced in a cell-or tissue-specific manner and are generally limited in their taxonomic distribution.
