Acute Hemodynamic Effects of Tetramethylpyrazine and Tetrandrine on Cirrhotic Rats

Abstract: The hemodynamic effects of tetramethylpyrazine (TMP) and tetrandrine (TET), both alkaloids isolated from Chinese herbs Ligusticum wallichii Franch and Stephania tetrandra S. Moore, respectively, were assessed in anesthetized cirrhotic rats. TMP induced dose-dependent decreases of portal venous pressure (P.V.P.) and mean arterial pressure (M.A.P.) after intravenous infusion. The maximum percentage reduction of P.V.P. after TMP was 3.6 +/- 0.8%, 6.8 +/- 0.5%, and 17.8 +/- 0.6% of baseline, respectively, for the … Show more

“…NAC, as a positive control, also inhibited the PDGF‐stimulated BrdU incorporation. TMP, a pure compound isolated from LC and previously demonstrated to have antiportal‐hypertensive or hepatoprotective effects, 17,18 did not exert inhibitory effects on PDGF‐induced BrdU incorporation even up to the concentration of 300 µmol/L. Taken together, these results suggested that TMP is not likely to be responsible for the inhibitory effects of the LC extract observed in the present study.…”

“…These indications are partially due to the vasodilatory, platelet‐inhibitory and antithrombotic effects of LC on patients 12,13 . Tetramethylpyrazine (TMP), an active compound of LC, has been reported to be an agent against platelet aggregation, 14 a vasodilator with calcium channel blocking activity, 15 a scavenger of superoxide anion in human polymorphonuclear leukocytes, 16 and a chemical with antiportal‐hypertensive and hepatoprotective effects 17,18 . However, the potential of LC as antifibrotic agent has not yet been explored.…”

“…Tetrandrine decreases of PVP and MAP, the maximum percentage reduction of PVP after drug was 5.4 ± 1.0%, 9.2 ± 0.8% and 23.7 ± 1.2% of base line, respectively, for the doses given 2.0, 6.6 and 20.0 mg/kg. Total peripheral resistance was also reduced by the drug (Huang et al, 1999).…”
