“…Thus, in the cotreated groups the elevations in 2,5-HD concentration observed in serum and the sciatic nerve most likely resulted from the diminished K e value. These results are supported by the decrease in 2,5-HD elimination reported on coexposure to 2,5-HD and MEK (Ralston et al 1985) or to 2,5-HD and AC (Ladefoged and Perbellini 1986). As most of the 2,5-HD is eliminated after being metabolized in the liver, the relationship between liver metabolic capacity and 2,5-HD concentration changes should be clari®ed.…”

“…A number of studies have been conducted since the late 1970s on the possible eects of co-exposed solvents on the metabolism of HEX (Altenkirch et al 1978, 1982a, b, Takeuchi et al 1981, Iwata et al 1984, Abou-Donia et al 1985, Ladefoged and Perbellini 1986, Shibata et al 1990a, b, Nylen and Hagman 1994, Nylen 1996, Ichihara et al 1998). The animals exposed were in most cases rats, and the solvents tested for combined eects included TOL, xylenes (XYL) and MEK.…”

“…However, a toxicokinetic interaction with biotransformation or renal excretion of 2,5-HD cannot be excluded. This possibility has recently been investigated in rabbits where it has been shown that acetone given together with 2,5-HD reduced the total body clearance of 2,5-HD (Ladefoged & Perbellini 1986). This mechanism may also partly explain the potentiation of the neurotoxicity of nhexane by the ketone MEK.…”
