ABO Blood-Typing Using an Antibody Array Technique Based on Surface Plasmon Resonance Imaging

Abstract: In this study, readily available antibodies that are used in standard agglutination tests were evaluated for their use in ABO blood typing by a surface plasmon resonance imaging (SPR imaging) technique. Five groups of antibodies, including mixed clones of anti-A, anti-B, and anti-AB, and single clones of anti-A and anti-B, were used to construct the five-line detection arrays using a multichannel flow cell in the SPR imager. The red blood cell (RBC) samples were applied to a multichannel flow cell that was ort… Show more

“…SPR immunoassay can also be performed in array technique, which has the ability to be used with high-throughput and low cost method (Houngkamhang et al, 2013). Although, there are no reports related to cardiac biomarker detection using SPR immunosensor in array technique so far, this method offered an advantage of testing parallel which several samples, in a single run (Houngkamhang et al, 2013).…”

“…This work introduces another novel application of SPRi in quantifying the density of surface antigens expressed by different cell lines [26]. A series of studies were performed in this area on different types of cell lines such as murine macrophages [37], or ABO blood-typing using different classes of antibodies [38]. These studies demonstrate the utility of SPR in conjunction with immune markers for the sensitive detection of cell populations that are present with relatively low concentration in whole blood.…”

“…Recently, various studies have been reported in the applications of SPR technique for the study of RBCs. Houngkamhang et al [27] reported ABO blood typing via SPR imaging which observed interaction between immobilized Anti-A and Anti-B antibodies array and A and B antigens on RBCs surface. The interaction of antigens around lower RBCs membrane surface and immobilized Anti-A and Anti-B antibodies array was applied to obtain the agglutination strength of the RBCs and immobilized antibody [28].…”
