A Validated LC-MS/MS Method for the Quantification of Trigonelline in Marketed Dietary Supplements

Abstract: Background: Fenugreek seeds are employed in many traditional systems as an antibacterial, antidiabetic agent, gastric stimulant, and also for anti-invasive activity. Therefore, it is a suitable bioactive marker to establish the quality of crude drug and its formulations. Methods: A rapid, simple and sensitive LC-MS/MS analytical method was developed and validated for the determination of trigonelline extracted from Trigonella foenum-graecum (L.) (Fenugreek) and marketed dietary supplements using Etofylline a… Show more

“…Intriguingly, three alkaloids were characterized in the studied B. armeniaca leaves, designated as trigonelline, harmaline, and caffeine. Trigonelline ( m / z 138.0551, peak 13) produced a fragment ion at m / z 92.0489 [(M + H)-C 2 H 6 O] derived from an ethyl unit loss (−46 Da) and CO neutral loss (−28 Da) indicated at m / z 110.0599 [ 57 ]. Meanwhile, harmaline (peak 17), with a molecular ion peak at m / z 215.1401, yielded main fragment ions at m / z 200.1342 [(M + H)-CH 3 ] and m / z 169.1384 [(M + H)-CO-H 2 O] [ 58 ], whereas caffeine (peak 28) displayed a pseudo molecular ion [M + H] + at m / z 195.1122 with a base peak at m / z 138.0005 [ 59 ].…”

“…Peak No. 11 was assigned to trigonelline (tr = 1.4326, min, [C 7 H 8 NO 2 ] + ), with the molecular ion 138.0522 and a fragment ion at m/z 92.0489 indicating the loss of an ethyl unit [M + H − C2H6O] + [40] (Figure S20). Caffeine (tr = 5.0098, min, [C 8 H 11 N 4 O 2 ] + ) was assigned to Peak No.…”
