The I-3 gene from the wild tomato species Lycopersicon pennellii confers resistance to race 3 of the devastating vascular wilt pathogen Fusarium oxysporum f. sp. lycopersici. As an initial step in a positional cloning strategy for the isolation of I-3, we converted restriction fragment length polymorphism and conserved orthologue set markers, known genes and a resistance gene analogue (RGA) mapping to the I-3 region into PCR-based sequence characterised amplified region (SCAR) and cleaved amplified polymorphic sequence (CAPS) markers. Additional PCR-based markers in the I-3 region were generated using the randomly amplified DNA fingerprinting (RAF) technique. SCAR, CAPS and RAF markers were used for high-resolution mapping around the I-3 locus. The I-3 gene was localised to a 0.3-cM region containing a RAF marker, eO6, and an RGA, RGA332. RGA332 was cloned and found to correspond to a putative pseudogene with at least two loss-of-function mutations. The predicted pseudogene belongs to the Toll interleukin-1 receptor-nucleotide-binding site-leucine-rich-repeat sub-class of plant disease resistance genes. Despite the presence of two RGA332 homologues in L. esculentum, DNA gel blot and PCR analysis suggests that no other homologues are present in lines carrying I-3 that could be alternative candidates for the gene.

Abstract. This study was carried out to establish the simple mass-screening methods for resistant tomato to Fusarium wilt caused by Fusarium oxysporum f. sp. lycopersici (FOL). Root dip inoculation method has been used in many studies on the resistance of tomato to disease. On the other hand, in mass-screening for resistant tomato to Fusarium wilt, the inoculation method is time-consuming and laborious procedure. Disease development of two FOL isolates on two cultivars of tomato according to inoculation method including root dip, tip and scalpel methods were investigated. In compatible interaction, tomato seedlings of each cultivar inoculated by tip method showed the lower and more variable disease severities than by root dip method. Whereas the seedlings by scalpel method represented clear resistant and susceptible responses to Fusarium wilt as root dip method. The resistance degree of each cultivar inoculated with FOL isolates by scalpel method was hardly affected by the tested incubation temperature and inoculum concentration. On the basis of the results, we suggest scalpel inoculation method as an efficient mass-screening method for resistant of tomato cultivars to Fusarium wilt. La marchitez por Fusarium causada por Fusarium oxysporum f. sp. lycopersici (Fol) es una de las enfermedades más importantes del cultivo del tomate (Solanum lycopersicum L.). Su variabilidad patogénica ha originado las razas 1, 2 y 3, ya descritas en varios países. En México se ha reportado en Sinaloa, Baja California Sur y Morelos. El tomate es un cultivo hortícola muy importante en Villa de Arista, San Luis Potosí, por lo que el objetivo de este trabajo fue caracterizar mediante pruebas de patogenicidad la variabilidad en la virulencia de aislamientos de Fol en predios tomateros de esta región. Se obtuvieron 27 aislamientos de Fusarium oxysporum de plantas de tomate con síntomas característicos de la marchitez por Fusarium de nueve variedades en siete predios tomateros, las cuales se procesaron en laboratorio para aislar el hongo, purif icarlo, preparar inoculo y realizar pruebas de patogenicidad. Las variedades diferenciales utilizadas fueron Bonny Best, susceptible a las razas 1, 2 y 3; Manapal, resistente a la raza 1; Walter, resistente a las razas 1 y 2; e I3R3, resistente a las razas 1, 2 y 3. Las razas identificadas fueron, raza 2 en las variedades Rafaello y Tipsey en los predios Agro Viva y Rancho el Clérigo, y la raza 3 en las variedades El Cid, Aníbal y 77-05 en los predios San Gilberto, Santa María Elena yLourdes.Laidentificacióndeestasrazasmediantelatécnica molecular de la Reacción en Cadena de la Polimerasa (PCR) coincidió con los resultados de las pruebas patogénicas.
