A substitute to xylene in deparaffinization and clearing prior to coverslipping in histopathology

Abstract: INTRODUCTION: Deparaffinization and clearing prior to coverslipping are important steps in all staining methods in histopathology. Xylene is the most commonly used agent worldwide. However, xylene is toxic. We evaluated safer alternative dewaxing and clearing agents prior to coverslipping in a histopathology laboratory. MATERIALS AND METHODS: Thirteen different fresh surgical tissues were cut into two halves. One half processed using xylene and the other half processed using UltraClear™. Five groups … Show more

“…The mixture is used in hand washing of cutlery. The concept of using dish wash liquid as dewaxing agent is because of its surfactant property reducing the surface tension [ 11 ]. Some have highlighted the advantages of hot dishwashing soap (DWS) solution for deparaffinization of tissue sections for H&E staining and stated that even in special staining procedures like periodic acid-Schiff (PAS) staining and Van Gieson staining it can be applied.…”

“…While the grinding of the embedded samples into small pieces was a procedure in common with similar toxicological research articles [11,12], the solubilization of the paraffin wax embedding the samples was a more delicate procedure. Classic deparaffinization involving the use of xylene or a xylene substitute and subsequent rehydrating passages in a decreasing alcoholic ladder [17] was considered as too invasive under a toxicological point of view; the washing out and subsequent loss of analytes had to be taken into consideration, and we therefore decided to proceed by solubilizing paraffin in a non-polar solvent n-hexane. Analytes of toxicological interest are polar molecules and we therefore decided to proceed with a liquid-liquid extraction using acetonitrile.…”
