A simple RP-HPLC method for the simultaneous determination of curcumin and its prodrug, curcumin didecanoate, in rat plasma and the application to pharmacokinetic study

Abstract: A high-performance liquid chromatographic (HPLC) method has been developed for the simultaneous determination of curcumin and its prodrug, curcumin didecanoate (CurDD), in rat plasma. The analytes were extracted by ethyl acetate following the addition of sodium dodecyl sulfate, and separated on a reverse-phase C(18) column using a gradient mobile phase system of acetonitrile-tetrahydrofuran-water containing 0.1% formic acid. Detection by UV absorption at 425 nm gave a lower limit of quantitation (LLOQ) of 5 an… Show more

“…Beevers and Huang (2011) determined the hydrolysis of curcumin diethyl disuccinate in human plasma and in phosphate buffer (pH 7.4) they followed pseudo first order kinetics. It also maintains plasma levels in humans (Han et al 2011). In one study conducted in Greece Pandelidou et al (2011) prepared a stable curcumin formulation that comprised egg phosphatidylcholine (EPC) liposomes.…”

“…A high-performance thin layer chromatography ( Different HPLC methods have been reported for the analysis of curcumin, such as HPLC methods with fluorescence detector have been used for its quantification in biological samples [10]. HPLC-UV methods were also used for the quantitative determination of curcumin in biological samples [11]. HPLC-PDA methods were also reported for determination of curcumin [12] and simultaneous estimation of piperine and curcumin in nanoparticulate formulation [13].…”

“…The reported methods were costly due to the use of highly sophisticated instruments like mass spectrometry [16] and detectors like fluorescence detectors [10]. The use of expensive solvents such as tetrahydrofuran [11,12] was replaced by acetonitrile in this study. The LOD and LOQ were found to be 0.018 and 0.056 μg/ml, respectively, which indicates that the method was sensitive, and can detect and quantify at lower levels of curcumin.…”

“…Various role including antioxidant, anti-inflammatory, antibacterial, antiviral, anti-atherosclerotic, anti-arthriticagent and anticancer activities [16][17][18][19] are logical reasons for its extensive application and encourage researchers to design an accurate approach for preconcentration and quantification of curcumin in low level in complicated matrices. Several methods such as high-performance liquid chromatography [20][21][22], thin layer chromatography [23], UV-Vis spectrophotometry [24], HPTLC [25], 2-D flash liquid chromatography [26] and spectrofluorimetry [27,28] are applied for this regard. HPLC method compared to other techniques benefit from remarks like high reliability and efficiency and selectivity is appropriate for analyzing real samples with complex matrices, while for analytes with extremely low concentration a preliminarily preconcentration help the researchers to reduce the interference correspond to other constituents and simultaneously improve the performance of proposed method [29].…”
