A retrospective study detects a novel variant of porcine epidemic diarrhea virus in England in archived material from the year 2000

Abstract: Outbreaks of porcine epidemic diarrhea (PED) were first recorded in England in the 1970s and continued to be confirmed until 2002. Retrospective analysis of archived material from one of the last confirmed cases in England in the year 2000 demonstrates the previous existence of a very diverse PED virus strain. Following the outbreaks of PED in North America in 2013, there has been renewed interest in phylogenetic analysis of sequences from PEDV strains worldwide. There is a gap in the available sequence data b… Show more

“…Since spring 2014, outbreaks of diarrhoea affecting pigs of all age groups have also been reported in farms in Europe, firstly in Germany (Henniger and Schwarz, 2014;Hanke et al, 2015;Stadler et al, 2015), followed by its neighbouring countries, the Netherlands, Belgium, and Austria (Theuns et al, 2015;Steinrigl et al, 2015), as well as from the more distant Ukraine in summer 2014 . However, only the outbreak in the Ukraine was caused by a Colorado-like PEDV strain, whereas all the central European outbreaks were caused by the Ohio851-like variant.…”

“…While most of the historic material held in our storage is from earlier passages of CV777, we also had material from one of the last GB PEDV cases in the year 2000 available for analysis (Steinbach et al, 2016). This PEDV strain, termed England-1-2000, is indeed significantly different not only to CV777, but also to all other previously known strains and can be seen as a prototype of a further genotype of PEDV (Fig.…”

“…NON-INDEL strains subsume USA/Kansas29/2013 strains (GenBank accession number KJ645637.1). These viruses caused a more severe disease course and higher mortality compared to the S-INDEL strains (17,18,25,31). All PEDV pandemic strains circulating in China after 2014 were also clustered to genotype 2, and they were genetically separated from other global PEDV strains and from earlier Chinese strains (29).…”

“…The N protein has multiple functions in viral replication, interacts with viral genomic RNA, and associates with other N proteins to protect the viral genome. It disturbs antiviral responses by antagonising interferon production (17,31). The S protein is encoded by a gene sequence which determines the virulence of a PEDV strain (35).…”

“…Within the groups a certain variety can be distinguished, hence subgroups of strains have been described, namely G1a and G1b and G2a and G2b. In addition, the strains belonging to PEDV group G2 which have a deletion or insertion in the S gene were classified as S-INDEL variants from the originally isolated CV777 strain ( 87 , 96 , 97 ). The virus may spread with traded pigs that are asymptomatically infected and shed the virus into the environment with faeces.…”
