A recommended esterification method for gas chromatographic measurement of conjugated linoleic acid

Abstract: The effect of the methylation method on isomerization of conjugated linoleic acid (CLA) in gas chromatographic analysis was studied. Among methylation methods examined, the magnitude of isomerization of CLA was greatest with BF 3 catalyst, followed by HCl and H 2 SO 4 catalyst. Shorttime methylation did not extensively change the CLA composition in all methods, and c,t and t,c isomers were essentially maintained, while the appearance of t,t isomers and unknown peaks was practically restricted. After 120 min of… Show more

“…c9,c11-CLA, c9,t11-CLA, t9,t11-CLA and t10,c12-CLA were purchased from Matreya, Inc. (State College, PA, USA). Each CLA was converted to corresponding methyl ester with H 2 SO 4 -methanol solution [16]. Each methyl ester was analyzed by capillary gas chromatography (GC).…”

“…Para definir el perfil lipídico de las especies de gramíneas, se realizó la extracción y metilación de los ácidos grasos adaptando las técnicas descritas por Garcés y Mancha (1993) y por Yamasaki et al (1999). Al forraje (50 mg) se le adicionaron 2.150 μl de metanol absoluto, 990 μl de tolueno, 66 μl de ácido sulfúrico al 99,9 %, 1.000 μl de N,N dimetilformamida y 2 ml de n-hexano.…”

“…For esterified lipids, artefacts are still produced (because more severe reaction conditions are required), but at a much lower quantity than those arising from the use of BF 3 . 45,59 Some authors showed good results using a traditional base-catalysed methylation, followed by a mild treatment using H 2 SO 4 -methanol. 45 In general, acid-catalysed methods can be used in a direct transesterification or in a two-step methylation incorporating a previous hydrolysis or saponification of the lipids to free fatty acids.…”
