A Novel and Effective Strategy for the Isolation of Adipose-Derived Stem Cells

Abstract: Adipose-derived stem cells are an ideal mesenchymal stem cell population for regenerative medical application. The isolation procedure is performed by mechanical isolation under a laminar air flow bench without using serum or animal-derived reagents; cells were characterized by flow cytometric analysis. Cell availability is improved compared with enzymatic digestion procedures. The adipose-derived stem cell mechanical isolating procedure presented here is easier, safer, cheaper, and faster than traditional cur… Show more

“…[76], [77], [78] described a method that was specifically designed for clinical application, which appeared easy, safe and fast (80 min), allowing collection of a ready-to-use ASC pellet. Just like previous isolation processes, first the adipose tissue was harvested by a standard liposuction procedure, and then the isolation process was carried out through both mechanical (centrifugation) and enzymatic (collagenase) means [78].…”

“…However, the clinical application of ASCs isolated using mechanical means during the same operative session falls under the jurisdiction of medical practice and is thus allowed. Therefore, the development of high-yield isolation processes for ASCs with minimal handling would be highly desirable for clinical applications [76].…”

“…Constricted skin with a very thin or scarred subcutaneous fat layer cannot sustain as large an amount of fat as healthy tissue, preferably with lax skin and a thick subcutaneous fat layer [88]. In fact, the vitality of ASCs has proven to be crucial for stimulating tissue regeneration through the chemotactic, paracrine, and immunomodulatory activities of the cells and in situ differentiation [38], [39].…”

“…Stem cells isolated from lipoaspirates have demonstrated a broad in vitro adipogenic, chondrogenic, osteogenic, and myogenic lineage commitment [33], [34] as well as differentiation into pancreatic cells, hepatocytes, and neurogenic cells [35], [36], [37]. Cytometric analysis of adipose-derived stem cells (ASCs) has shown that these cells do not express CD31 and CD45, but do express CD34, CD73, CD105, and the mesenchymal stem cell marker CD90 [38], [39]. ASCs have a differentiation potential similar to that of other mesenchymal stem cells as well as a higher yield upon isolation and a greater proliferative rate in culture than bone marrow–derived stem cells [40], [41], [42].…”

“…Newer methods claim quicker and cheaper isolation [40], and some attempt to bypass enzyme digestion all together [53]. Because a key step in adipose-derived stem cell retrieval is the isolation of the SVF (the pellet of mixed cell types after centrifugation of collagenase-treated lipoaspirate), many commercial devices have surfaced over the last decade targeting SVF isolation.…”
