A New Cytotoxicβ-Carboline Alkaloid fromGalianthe thalictroides

Abstract: A cytotoxicity-guided fractionation of the roots of Galianthe thalictroides afforded a new β-carboline alkaloid, 1-(hydroxymethyl)-3-(2-hydroxypropan-2-yl)-2-(5-methoxy-9H- β-carbolin-1-yl)cyclopentanol, which exhibited strong cytotoxic activity against three human cancer cell lines (MCF-7, 786-0, and UACC62). Its structure was established on the basis of 1D- and 2D-NMR spectroscopic techniques supported by HRMS data.

“…These data agree with the results of Figueiredo et al (2011), in which the same CHCl 3 phase exhibited a high cytotoxic activity against MCF-7 Table 5 Frequency of mutant spots in the wings of marked trans-heterozygous descendants (mwh/flr 3 ) of D. melanogaster using the standard crossing (ST) and high bioactivation cross (HB) after chronic treatment with the ethanol extract (EtOH) of G. thalictroides. “…This difference in toxicity should be attributed to the presence of other metabolites in the bioactive CHCl 3 phase. In a previous work, differences between the cytotoxic behavior of this phase and alkaloid 1 were also observed against MCF-7 cell lines (Figueiredo et al, 2011). The recombination events induced by the CHCl 3 phase in flies from the cross were however not observed for alkaloid 1, and therefore the effects caused by this compound are shown to be exclusively related to its mutagenic activity, and since they were proportional to the concentrations applied, these results were indicative of a dose-response relationship.…”
