A human tissue‐engineered vascular media: a new model for pharmacological studies of contractile responses

Abstract: Our method for producing tissue-engineered blood vessels based exclusively on the use of human cells, i.e., without artificial scaffolding, has previously been described (1). In this report, a tissue-engineered vascular media (TEVM) was specifically produced for pharmacological studies from cultured human vascular smooth muscle cells (VSMC). The VSMC displayed a differentiated phenotype as demonstrated by the re-expression of VSMC-specific markers and actual tissue contraction in response to physiological stim… Show more

“…A response was recorded for seven of the nine drugs used; no response was measured for SNP or BHT-920. The number of drugs tested in this study is surpassed only by the study of cell sheet TEVMs by L'Heureux et al 6 The list of agents studied is significant, but not all-encompassing. Even if a response was found to all of the agents, it would not have proven that the cells were fully functional.…”

“…Cell sheet tissue-engineered vascular medias (TEVM) have been extensively tested through use of a comprehensive panel of agents, 6 as well as investigations of the endothelin system. 7 Despite issues with cell culture, vasoactivity has been seen in TEVM that used adult smooth muscle cells [8][9][10] as well as TEVM using umbilical cord derived, 6,7,11,12 bone marrow progenitor, 13 and neonatal cells. 14,15 One reason why few groups have demonstrated function is that obtaining the number of cells necessary to make a TEVM requires extensive passaging of cells.…”
