Abstract. During mammalian sex determination of XY fetuses, SRY induces SOX9 in Sertoli cells, resulting in formation of testes with seminiferous tubules, interstitial Leydig cells and peritubular myoid cells. Meanwhile XX fetuses without SRY develop ovaries. In cattle, most XX heifers born with a male twin, so-called freemartins, develop nonfunctioning ovaries and genitalia with an intersex phenotype. Interestingly, freemartins sometimes develop highly masculinized gonads with seminiferous tubule-like structures despite the absence of SRY. However, in these cases, the degree of masculinization in each gonadal somatic cell type is unclear. Here, we report a rare case of a freemartin Japanese black calf with almost complete XX sexreversal. Gross anatomical analysis of this calf revealed the presence of a pair of small testis-like gonads with rudimentary epididymides, in addition to highly masculinized genitalia including a pampiniform plexus, scrotum and vesicular gland. Histological and immunohistochemical analyses of these masculinized gonads revealed well-defined seminiferous tubulelike structures throughout the whole gonadal parenchyma. In epithelia of these tubules, SOX9-positive supporting cells (i.e., Sertoli cells) were found to be arranged regularly along the bases of tubules, and they were also positive for GDNF, one of the major factors for spermatogenesis. 3β-HSD-positive cells (i.e., Leydig cells) and SMA-positive peritubular myoid cells were also identified around tubules. Therefore, for the first time, we found the transdifferentiation of ovarian somatic cells into all testicular somatic cell types in the XX freemartin gonads. These data strongly support the idea of a high sexual plasticity in the ovarian somatic cells of mammalian gonads.

This paper reports the evaluation of anorectal malformations with two different fistula in two calves. During clinical examination atresia ani and meconium coming from preputium was observed in both calves. Distention of intestines was viewed in radiological examination. Surgical treatment had been preferred in both calves. During surgery it was observed that colon was connected to the bladder with tubelike formation in calf 1. In calf 2 colo-urethral fistula was detected during necropsy. The clinical, surgical and necropsy findings of anorectal malformations with colovesical and colourethral fistula in two calves has been reported for the first time. As a conclusion, the localization of the fistula and the condition of the animal could affect results of surgical treatment. Her iki buzağının klinik muayenesi sırasında atresia ani ve mekonyumun prepisyumdan geldiği belirlendi. Radyografik muayenede ise bağırsakların genişlediği gözlendi. Her iki buzağının opeartif tedavisine karar verildi. Birinci buzağının operasyonunda kolonun tüp şeklinde bir yapı ile idrar kesesine bağlandığı belirlendiği. İkinci buzağıda ise kolonun urertraya fistülleştiği nekropsi sırasında anlaşıldı. Kolovesikal ve koloüretral fistüle sahip iki buzağının klinik, cerrahi ve nekropsi bulguları ilk defa sunulmaktadır. Sonuç olarak, fistülün lokalizasyonu ve hayvanın genel durumu cerrahi tedavinin sonucunu etkileyebilmektedir.

ÖzetFreemartinismus sendromu, farklı cinsiyette yavruların bulunduğu ikiz gebeliklerde erkek yavruya ait kan kök hücrelerinin ve anti-Müllerian faktörü hormonunun dişi ikizin dolaşımına karışması sonucu dişi yavruların steril olması durumudur. Erkek ikizi olan dişi buzağılarda %82.5 ve %92 oranında freemartinusmus gelişebilir. Freemartin bireylerin belirlenmesinde çeşitli yöntemler kullanılmaktadır. Ancak freemartin yavruların erken dönemde belirlenmesi, yavrunun damızlık olarak kullanılıp kullanılmaması için önemlidir. Erken dönemde yavrunun freemartin olup, olmadığının belirlenmesi sitogenetik veya Y spesifik primerlerin kullanıldığı moleküler yöntemlerle yapılabilir. Ancak hassasiyeti, uygulama kolaylığı ve tanı süresinin kısalığı yönünden moleküler yöntemler tercih edilir. Bu çalışmada, erkek ikizi olan üç dişi buzağının freemartinismus durumları polimeraz zincir reaksiyonu (PZR) ile araştırılmıştır. PZR işlemi Y kromozomuna özel iki primer seti kullanılarak yapılmıştır. Ayrıca bu yavruların vajina uzunlukları ve internal üreme kanalı transrektal ultrason ile incelenmiştir. PZR işlemi sonunda erkek ikiz kardeşi olan dişi yavruların ikisinin Y kromozomuna sahip olduğu, bir buzağının ise Y kromozomu taşımadığı belirlenmiştir. Aynı zamanda bu yavrunun vajina uzunluğunun incelenen diğer buzağılardan daha uzun olduğu (9 cm) ve ultrasonografik incelemeler sonunda serviksin bulunduğu görülmüştür. Bu çalışmadan elde edilen bulgulara göre erkek ikizi olan dişi yavruların ve tekrarlayan tohumlamalarda gebe kalmayan damızlık dişi sığırların freemartinismus yönünden araştırılmasında PZR yönteminin kullanılabileceği sonucuna varılmıştır. Freemartinism may develop in female twin at a rate of 82.5% and 92% in twins with diff erent sexes. Diff erent methods are used to determine freemartinism syndrome. However, early identification of freemartinism is very important to determine the potential of calf for breeding purposes. Early identification of freemartinism could be accomplished by cytogenetic and molecular methods via Y-specific primers. However, molecular methods are preferred due to their accuracy, ease of application and shorter duration of process. In this study, freemartinism status of three female calves born with twin male calves was investigated by polymerase chain reaction (PCR). PCR process was carried out by using two primer sets, specific to Y-chromosome. Moreover, the length of the vagina and reproductive tract was examined by transrectal ultrasonography.
