9‐cis Retinoic acid induces the expression of the uncoupling protein‐2 gene in brown adipocytes

Abstract: The expression of uncoupling protein-2 (UCP2) mRNA is up-regulated during the differentiation of brown adipocytes in primary culture. When differentiation of brown adipocytes is impaired, UCP2 mRNA expression is downregulated. 9-cis Retinoic acid causes a dose-dependent induction of UCP2 mRNA levels in brown adipocytes, whereas all-trans retinoic acid has no effect. Specific agonists of retinoid X receptors (RXR) induce UCP2 mRNA expression, whereas specific activators of retinoic acid receptors do not. 9-cis … Show more

“…Gene expression in what parts of animals was related to its corresponding gene function. RXR bound with 9-cisRA induced expression of UCP2 gene to promote brown fat cell differentiation in adipose tissue [11]. It was indicated that RXR together with PPAR in adipocyte regulated the differentiation of preadipocyte [12].…”

“…There was only one report about expression profile of the cattle embryo [10], however, the integrated expression profile of bovine RXR gene is still vacant so far. In functions, Carmona et al [11] reported RXR by binding 9-cisRA induced expression of UCP2 gene to promote brown fat cell differentiation. The previous research indicated RXR, together with PPAR to form PPAR/RXR heterodimer in adipocyte, regulated the differentiation of preadipocyte [12].…”

“…Moreover, evidence was provided that PPARg does not bind directly to the mouse UCP2 gene promoter [59]. In spite of that, retinoids [46,60,61] and PPAR ligands ( [59] and references therein) have been reported to induce UCP2 gene expression in different systems. Focusing on retinoids, 9-cis RA was shown to upregulate UCP2 mRNA expression in brown adipocytes in primary culture [60], and all-trans-RA was shown to do the same in L6 myotubes [61].…”

“…Retinoic acid (RA), its carboxylic acid form, promotes (9) or inhibits (10) adipogenesis of preadipose cells in culture depending on the dose, negatively affects preadipocyte survival (11), and induces the expression of uncoupling proteins (UCPs) in cultured brown adipocytes (12)(13)(14). Moreover, both RA treatment and vitamin A status influence body adiposity and the expression of adipogenic/ lipogenic transcription factors and UCPs in white (WAT) and brown (BAT) adipose tissues and of UCP3 in muscle of rodents (13,(15)(16)(17).…”
