4T1 Mammary Carcinoma Colonization of Metastatic Niches Is Accelerated by Obesity

Abstract: Breast cancer (BC) remains the leading cause of cancer-related deaths among women, and the chances to develop it are duplicated by obesity. Still, the impact of obesity during BC progression remains less understood. We investigated the role of obesity in tumor progression using the murine model of 4T1 mammary carcinoma in BALB/c female mice, previously high-fat-diet (HFD) fed. HFD induced obesity, metabolic impairment, and high serum and fat leptin levels. After injection of 4T1-cells, HFD-mice accelerated tum… Show more

“…Consumption of a high‐fat diet increased the growth rate and size of primary tumors in the 4T1 BALB/c model, regardless of dietary pattern. This matches trends found in other 4T1‐high fat diet studies (Evangelista et al, 2019 ; Kim et al, 2011 ); though it is notable the source of fat calories varies between these reports. Western diet was notably the most detrimental, leading to the highest rate of tumor growth and the largest tumor size.…”

“…11 The effect of obesity does not end with this; it also results in breast and ovarian cancers. 12 The risk of obesity was higher among users of hormonal contraceptives due to the effect of progesterone. Progesterone induces appetite or facilitates anabolism and oestrogen aggravates the accumulation of fat in the cell or adipose tissue.…”

“…Obesity leads to development of different cancers; this is why, cancer epidemiology abruptly increased worldwide. Obesity also promotes breast cancer formation [77] and formation of Barrett's esophagus (BE) [78]. BE has been defined as a pathological state in which the stratified squamous epithelium of the distal esophagus has been replaced by the metaplastic columnar epithelium with goblet cells.…”
