Protective effect of alpha-lipoic acid in methotrexate-induced ovarian oxidative injury and decreased ovarian reserve in rats

Abstract: To determine whether the possible oxidative effect of methotrexate (Mtx) on ovary and to evaluate the effectiveness of alpha lipoic acid (ALA), which may be useful in many oxidative stress models. Thirty-two female Wistar-albino rats were randomly divided into four groups; control group, alpha lipoic acid group (ALA 100 mg/kg, 10 days), multiple dose Mtx group (Mtx 1 mg/kg 1, 3, 5, 7 days) and Mtx and ALA group (Mtx 1 mg/kg 1, 3, 5, 7 days and ALA 100 mg/kg, 10 days). Serum total antioxidant status (TAS), tota… Show more

“…The etiology and pathogenesis of DOR remain unclear so far, but it has been reported that DOR may be related to age (Fan et al, 2019), immunity (Silva et al, 2014), heredity, environment, radiotherapy and chemotherapy (Smith et al, 2013), surgery, psychological stress (Nicoloro-SantaBarbara et al, 2017), and infectious factors (Soylu Karapinar et al, 2017). A prospective cohort study found that poor ovarian response was related to age and apoptosis of ovarian granulosa cells (Fan et al, 2019).…”

“…Similar to ours, in a study where instead of DHEA, alpha-lipoic acid utilized; it reported that in rats that received MTX, alpha-lipoic acid has reversed the AMH levels and the total follicle counts, and thus healing the oxidative damage to the ovaries. Karapınar and colleagues have concluded that the oxidative effects of MTX on the ovaries may be inhibited by alpha-lipoic acid supplementation which is an antioxidant (Soylu Karapinar, Pinar, Özcan, Özgür, & Dolapçıoğlu, 2017).…”

“…The oxidative stress is deemed to be one of the dominating mechanisms of ovarian failure (Devine et al., 2012; Luderer, 2014). It has been reported that intragastric or intraperitoneal administration of ALA improves oxidative damage to the ovaries (Avci, Bahadir, Bahadir, Tuncel, & Bilgici, 2016; Soylu Karapinar, Pinar, Pinar, Ozcan, Ozgur, & Dolapcioglu, 2017). In this study, 600 mg/kg ALA‐treated group decreased the level of MDA and improved the activity of TAOC and SOD in ovary.…”
