Amnion, which is usually discarded as medical waste, is considered as abundant sources for mesenchymal stem cells. In human and veterinary medicine, the multipotency of mesenchymal stem cells derived from amnion (AMSCs) together with their plasticity, self-renewal, low immunogenicity and nontumorigenicity characteristics make AMSCs a promising candidate cell for cell-based therapies and tissue engineering. However, up till now, the multipotential characteristics and therapeutic potential of AMSCs on preclinical studies remain uncertain. In this work, we successfully obtained AMSCs from Beijing duck embryos in vitro, and also attempted to detect their biological characteristics. The isolated AMSCs were phenotypically identified, the growth kinetics and karyotype were tested. Also, the cells were positive for MSCs-related markers (CD29, CD71, CD105, CD166, Vimentin and Fibronection), while the expression of CD34 and CD45 were undetectable. Additionally, AMSCs also expressed the pluripotent marker gene OCT4. Particularly, when appropriately induced, AMSCs could be induced to trans-differentiate into adipocytes, osteoblasts, chondrocytes and neurocytes in vitro. Together, these results demonstrated that the isolated AMSCs maintained their stemness and proliferation in vitro, which may be useful for future cell therapy in regenerative medicine.

Umbilical cord mesenchymal stem cells (UMSCs) from fetal tissues have become a valuable source of cell regeneration therapy due to their low immunogenicity, high plasticity and antimicrobial peptides. Most researches about UMSCs mainly focused on humans, mice, rabbits and other mammals, but it is still unknown in broiler. To the best of our knowledge, we first established a broiler-derived UMSCs line from noncontroversial Wharton's jelly of umbilical cord. The detection of biological characteristics showed UMSCs showed the morphological characteristics of fibroblasts and could be cultured to 26 generations in vitro, and maintained diploid chromosomes (2n=78) by G-banding analysis. Cell cycle of UMSCs showed the rate of G0/G1 was about 76.7-80.9%. Moreover, UMSCs could express mesenchymal markers (CD29, CD90, CD71, and CD44), but not hematopoietic marker (CD34) and endothelial marker (CD31). Additionally, UMSCs could be efficiently induced to transdifferentiate into three germ layer cells, such as adipocytes, osteoblasts, chondrocytes, hepatocytes, epithelia, and neurogenic cells. These results demonstrated UMSCs have the essential characteristics of MSCs, and can be used as an attractive candidate for cell regenerative therapy and the study of regulatory mechanisms associated with selective differentiation.

The role of mesenchymal stem cells (MSCs) in cellular therapy is well recognized in this work. MSCs have advantages of high proliferation, clone formation, multi-lineage differentiation and immunosuppression. Furthermore, adipose-resident MSCs (ADSCs) are extensively employed due to its advantages of abundant source, low cost and simple operation. Many researchers have emphasized the role of adipose-resident MSCs in the development of therapies for liver injury, but few attentions were paid on the use of induced functional hepatocytes. Therefore, in this work the role of adipose-resident MSCs induced functional hepatocytes was mainly investigated. The function of induced hepatocytes by ELISA and the induction rate was confirmed by flow cytometry and evaluated by experimental observations. The induced hepatocytes were firstly transplanted into CCl-caused liver damage ICR mice by tail vein. After transplantation, both liver fibrosis and function could be improved by hepatocytes, which were examined through histology, immunofluorescence staining, serum profile and biochemical parameters levels. The production of cytokines was then compared with normal mice and injury mice to explore the therapeutic mechanisms of hepatocytes. Finally, the secretions of TGF-β1, IL-6 and IL-10 in hepatocytes transplanted mice were determined and found to be higher than that of the normal and injury mice. The hepatocytes derived from ADSCs were proven to have a great significance in the therapeutic efficacy and clinical settings of liver disease animal models.
