Activation strategies for invariant natural killer T cells

Abstract: Invariant natural killer T (iNKT) cells are a specialized T cell subset that plays an important role in host defense, orchestrating both innate and adaptive immune effector responses against a variety of microbes. Specific microbial lipids and mammalian self lipids displayed by the antigen-presenting molecule CD1d can activate iNKT cells through their semi-invariant αβ T cell receptors (TCRs). iNKT cells also constitutively express receptors for inflammatory cytokines typically secreted by antigen-presenting c… Show more

“…A leaky gut barrier allows bacterial translocation, which is the escape of gut bacteria and their products through the intestinal mucosa to the outside of the intestine via portal or systemic circulation. This translocation is considered pathogenic in patients with chronic liver diseases who fail to remove bacteria or bacterial products, which leads to the accumulation of pathogen-associated molecular patterns that are recognized by Toll-like receptors in the liver and contribute to host’s immune system as well as liver damage and diseases in a chronic setting (104). Chronic alcohol administration leads to bacterial overgrowth along almost the entire gastrointestinal tract as well as dysbiosis characterized by reductions in probiotic bacteria such as Lactobacillus , Pediococcus , Leuconostoc , and Lactococcus (105).…”

“…Type I NKT cells, or invariant NKT (iNKT) cells, are characterized by the expression of a semi-invariant TCR composed of a Vα24Jα18 chain and a Vβ11 chain in humans, or a Vα14Jα18 chain paired with a limited repertoire of Vβ chains in mice [7,8,9,10]. …”
